The breakpoints of Df(2R)BSC879 predicted from the Release 5 genomic coordinates of the progenitor P{XP}Ef1alpha48Dd10427 and PBac{RB}CG8378e02998 transposable element insertion sites are 2R:7779605 ;8029867 and the cytological breakpoints predicted from these coordinates are 48C5;48E2.

The dorsal trunk of the tracheal system in Df(2R)reb-64/Df(2R)BSC879 embryos is curly, slightly irregular and elongated compared to controls. Despite its change in size, the tracheal network has normal branching patterns.

Lethal in combination with Df(2R)BSC199.

Df(2R)BSC329/Df(2R)BSC879 embryos show mild defects in chitin deposition - chitin accumulation in the dorsal trunk at stage 13 is delayed and later chitin levels in the dorsal trunk are slightly lower than wild type.

Df(2R)BSC879/Df(2R)ED2247 embryos do not accumulate the tracheal chitin filament and the trachea remain uninflated. A normal pattern of migration and tracheal cell specification is observed, but branches completely lack luminal and apical-cuticular chitin. General apicobasal polarity and adhesion are not affected. The dorsal trunk has a cystic appearance, with many dilations and constrictions, and the presence of apical expansions, usually at joints between branches; there are also abnormal cell shapes in the dorsal trunk.

Df(2R)BSC879/Df(2R)ED2247 embryos show a fragmented and granular head skeleton and an inflated and misshapen larval shape. The tracheal envelope and epicuticle is detached from the epithelial cells, while the space between these structures and the epithelial cells is devoid of material. The envelope and the epicuticle of the integument is decreased in thickness and the procuticle is amorph.

The presence of P+PBac{XP5.RB3}BSC879 was verified using the PCR methods and primers described in FBrf0175003 with the substitution of the primer 5’-GCTTCTAAACGCTTACGCATAAACGATG-3’ for the RB3’ plus or RB3’ minus primer in the Hybrid PCR protocol in the Supplementary Methods.
