Many fu41 mutant germaria contain increased numbers of spectrosome-containing GSC-like cells. Approximately 10% of germaria contain the normal number of GSC-like cells (<5), nearly 60% contain 5-10 cells and 30% of germaria are tumorous, containing in excess of 10 cells.

fu41 mutants exhibit a 'fused' wing phenotype, with fusion observed between L3 and L4.

In 4 day old fu41 homozygous females, only 7% of ovarioles are tumorous, but this has increased to 19% by 10 days. Tumorous follicles in these ovarioles consist of tens to hundreds of germ-line cells with small non-polyploid nuclei enveloped by a regular follicular epithelium. No increase in the rate of cell division of germline stem cells in the stem cell niche of the germarium is seen in these animals.

Mutants show fusion of wing veins L3 and L4 both proximally and distally, with intervein tissue remaining between the two veins in the medial part of the wing.

Mutant embryos die before hatching with mirror-image duplication of the abdominal denticle belts. Dorsal cuticle is mostly covered by a lawn of type 4 fine hairs. Cells type 1 are missing. The extent of naked cuticle (type 2 cells) is greatly reduced. Cells type 3 are usually present but sometimes missing. Type 4 cells are unaffected.

Does not suppress the phenotype produced by ectopic expression of hhScer\UAS.cCa using Scer\GAL430A.

When in combination with Su(fu)+ individuals exhibit a partially suppressed fu wing phenotype. Loss of one copy of Su(fu)+ causes a cos phenotype. fu41/fuK33R individuals display a strong fu wing phenotype, loss of proximal wing vein L4.

Heterozygotes carrying one copy of Su(fu)+ are pharate adult lethal and display a cos wing phenotype. The presence of P{SUFU100} or P{SUFU200} in these individuals produces viable males with a fu41 wing phenotype, the transposons fully compensate the decreased amount of Su(fu)+ product. With P{SUFU000} no males are recovered, consistent with lack of Su(fu)+ function. Heterozygotes carrying two copies are viable and display a fu wing phenotype. When the number of Su(fu)+ doses (via P{SUFU100}) in males is greater than two viability decreases, the decrease in viability is correlated with an enhancement of the fu wing phenotype. Decrease in viability is more pronounced when the transgene is provided by the mother. Viability of fu41 males is also affected when the Su(fu)+ activity decreases, the decrease in viability is associated with an anterior wing duplication phenotype (similar to cos mutant phenotype).

fu41 mutant embryos have pattern deletions in the posterior part of each segment in the cuticle, which are replaced by mirror duplications of the anterior part. The phenotype of fu41 ftz13, fu41 enLI034, and fu41 Ubx9.22 double mutants can be interpreted as an addition of the corresponding single mutant phenotypes. The phenotype of fu41 h25 double mutants can be interpreted as an addition of the corresponding single mutant phenotypes, except that the naked cuticle of segments A5 and A7 which is absent in the h25 single mutant, is replaced by a mirror duplication of the remaining denticle belts in the fu h double mutant. The ventral cuticle pattern is nearly wild type in fu41 nkd2 double mutants.

Anomalies of adult cuticular structures and tumorous ovaries.

strong wing phenotype - veins L3 and L4 fused all along length; intermediate viability phenotype - ratio of the number of fused flies to wild type in fu/+ X fu/Y is 0.5-0.75; weak fecundity phenotype - ratio of eggs laid by fu/fu versus fu/+ sisters is 0.5-1.0; strong maternal effect on segmentation - percent of embryos with all segments entirely duplicated is 70-100%.

Strong embryonic phenotype: the posterior portion of each segment is deleted and replaced with a mirror-image duplication of the anterior portion. Embryos die just before eclosion.

Expression of DadScer\UAS.P\T.cJa under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 suppresses the increase in spectrosome-containing GSC-like cells seen in fu41 mutant germaria.

Expression of tkvca.bam.T:Avic\GFP enhances the increased number of spectrosome-containing GSC-like cells seen in fu41 mutant germaria.

Homozygous lack15C has no effect on the increased number of spectrosome-containing GSC-like cells seen in fu41 mutant germaria.

In escaper flies that express Su(fu)Scer\UAS.cDa, under the control of Scer\GAL4da.G32, in a fu41 background, the wing phenotype is enhanced compared to flies expressing the transgene in a wild-type background. fu41, Su(fu)Scer\UAS.cDa, Scer\GAL4da.G32 wings show anterior duplications but also have a truncated vein 2, an enlargement of the domain between vein 2 and the margin, and veins 3 and 4 are completely fused with a large delta at the margin. These flies also show an enhancement of the leg phenotype seen in Su(fu)Scer\UAS.cDa, Scer\GAL4da.G32 flies.

fu41; Su(fu)LP/+ wing imaginal discs show overgrowth of the anterior compartment. fu41; ciCe-4/+ flies generally die as pupae, although some adults eclose. The latter have small wings, a reduced thorax and lack the scutellum. Partially suppresses the wing phenotype of hhMrt heterozygotes.

When in combination with Su(fu)LP, Su(fu)12d, cos3, Df(3R)kar3Q or Df(3R)kar-Sz11 heterozygotes individuals die as pupae. ptc mutations do not suppress the fu phenotype, fu1; ptc9/ptc9 embryos show no naked cuticle.

Complementation and Rescue Data

Comments

Expression of fu+t5.1 rescues the increase in GSC-like cells seen in fu41 mutant germaria.

Expression of fuKD fails to rescue the increase in GSC-like cells seen in fu41 mutant germaria.

Wing and embryonic phenotypes rescued by fu+t5.1 and fu+t7.4. Complete maternal rescue, 90% of embryos hatch to larvae and of these larvae 70% reach adulthood and those carrying fu+t5.1 or fu+t7.4 have wild type wings.

Mutant phenotype has been rescued by the introduction of the wild type fu gene by P element mediated transformation.

Mutant phenotype is partially rescued by injected fuCosC4 and less so by injected fucMa.
