eya2/eya2 and almost all eya2/eyacs adults are eyeless. eya2/eya4 adults exhibit smaller eyes compared to controls.

eya2/eya4 and eya2/eya3 adults exhibit significantly higher number of ommatidia in the eye than eya2/eyaE1, eya2/eyaE4, eya2/eya54C2 and eya2/eyaD3 adults.

eya2/eya4 but not eya2/eya3 adults exhibit significantly higher number of ommatidia in the eye than eya2/eyacli-IID adults.

eyacs/eya2 adults exhibit more severe eye phenotypes than eyacs/eya1, eyacs/In(2LR)ETD2.2, eyacs/eyacs and eyacs/eyaE(P)10 adults.

Adult eya2 homozygous mutants are eyeless.

Homozygous adults completely lack eyes.

Homozygotes show complete loss of eyes.

The eyes do not develop in flies carrying homozygous eya2.

Mutant flies show mild but significant defects in the auditory response to sound compared to wild type.

Instead of forming an even plexus of growth cones at the lamina, photoreceptor axons in eyaA188/eya2 larvae fasciculate aberrantly to produce an irregular pattern of gaps and thickenings in the lamina, with a significant percentage of axon bundles failing to terminate properly at the lamina.

The eclosion of eya2 mutant flies under photoperiod conditions lacks the prominent lights-on response seen in wild-type flies. No significant increases in eclosion are seen regardless of when flies are exposed to the lights-on signal. The eclosion rate is variable and substantial eclosion precedes the lights-on signal, but overall eclosion appears circadian. Flies exhibit normal geotaxis. The time taken for the flies to spread their wings post-eclosion is similar in flies that eclose in the dark and those that eclose in response to the lights-on signal. This is in contrast to controls, in which inflation occurs more quickly in the dark.

eya2 mutants have a small eye disc and show a complete loss of the retina.

Mutants completely lack eye tissue.

In eya2 embryos the distal tip of the salivary gland is not elongated along the anterior-posterior axis like in wild type, but is either oriented ventrally or curled underneath the long axis of the gland.

eya2 mutant flies have no ommatidia.

Homozygous mutant larvae show no glial cell migration defects in the developing eye.

Third instar larvae lack the developing adult photoreceptors. The dendritic arborisation of the lateral neurons appears normal.

Homozygotes completely lack eyes, whereas the ocelli are normal.

eya2/eya1 flies are eyeless. eya2/eya3 flies have eyes that are between 3/4 and less than normal size, and eya2/eya3 females are fertile.

eya2/eyaE3 flies are viable, and have eyes which range from 1/2 to less than normal size. eya2/eya4 flies have eyes which range from 1/2 to less than normal size. Homozygotes are eyeless. Homozygous clones in the eye do not survive. This phenotype is cell autonomous.

Homozygotes are completely eyeless. Eyes are restored to up to 3/4 of normal size in eya2 flies carrying one copy of eyaScer\UAS.cBa expressed under the control of either Scer\GAL4ey.PB, Scer\GAL4dpp.blk1, Scer\GAL4T59 or Scer\GAL4T155.

Homozygotes are completely eyeless.

Displays locomotor activity rhythm with an approximately 24h period.

Migration of the retinal basal glia from the optic stalk to the eye disc does not occur.

Homozygous flies are eyeless. eya2/eya4 heterozygous flies have 3/4 sized eyes.

Genetic Interactions

The eyeless phenotype of eya2 adult mutants can be partially rescued by expression of soScer\UAS.FL.NT.T:Hsim\VP16 (but not soScer\UAS.cUa) under the control of Scer\GAL4ey.PU in the mutant background.

Flies carrying homozygous eya2 and DNaseIIlo mutations die at the pupal stage.

Coexpression of AblK417N.Scer\UAS reduces the efficiency of eyaWT.Scer\UAS to rescue the 'eyeless' phenotype of eya2 from 100% to 40%.

Flies expressing eyScer\UAS.cHa under the control of Scer\GAL4dpp.blk1 in a eya2 background do not develop ectopic eyes, in contrast to flies expressing eyScer\UAS.cHa under the control of Scer\GAL4dpp.blk1 in a wild-type background.

Xenogenetic Interactions
Statement
Reference

Mmus\Eya2Scer\UAS.cBa restores eye formation in eya2 homozygotes when expressed under the control of Scer\GAL4ey.PB. The eyes produced have a pattern of ommatidial units with bristle cells similar to wild-type, and the pattern of photoreceptor neurons is normal.

eya2 is partially rescued by eyaRB.enh1

eya2 is partially rescued by eyaRB.enhE

eya2 is partially rescued by eyaRB.enh1+E

eya2 is partially rescued by eyaRB.enh1+s+2

Comments

The no-eye phenotype of adult eya2 homozygous mutants can be fully rescued by combination with eyaRB.enh1+E+2 and partially restored by combination with any of the following: eyaRB.enh1, eyaRB.enhE, eyaRB.enh1+E or eyaRB.enh1+s+2 but cannot be rescued by any of these transgenes: eyaRB.enh2, eyaRB.enh3, eyaRB.enh4, eyaRB.enh1+2, eyaRB.enh1+5bp+2 and eyaRB.+535.+840.

One copy of either eya+tGR, eyaD493N.GR, eyaE728Q.GR or eyaNQ.GR rescues rescues eye formation in eya2 homozygotes. The external and internal morphology of the eyes is indistinguishable from wild type.

One copy of eya+tGR fully rescues eye formation in eya2 homozygotes.

One copy of either eyaSA.GR or eyaSDE.GR rescues eye formation in eya2 homozygotes. The external morphology of the eyes are indistinguishable from wild type and cross sections show a normal number and arrangement of photoreceptors.

Inducible expression of eyaY4.Hsp70Bb from egg to pupa under the control of both Scer\GAL4hs.PB and Scer\GAL80ts.αTub84B rescues the eyes absent phenotype of eya2 mutants.

Inducible expression of eyaHsp70Bb.PL from egg to pupa under the control of both Scer\GAL4hs.PB and Scer\GAL80ts.αTub84B rescues the eyes absent phenotype of eya2 mutants.

The posterior and lateral regions of the eye are rescued by eyaScer\UAS.cPa expressed under the control of Scer\GAL4dpp.blk1. The development of the neuronal array is rescued in the most posterior region of the eye disc by eyaScer\UAS.cPa expressed under the control of Scer\GAL4E132.
