Overexpression of dppScer\UAS.cNa driven by Scer\GAL4dpp.PS promotes germ line stem cell proliferation in both young and aged germaria. There is a significant increase of egg production at day 3 after eclosion as a result of dppScer\UAS.cNa overexpression. However, the number of eggs produced by these females rapidly drops to comparable levels as that of the control flies at day 15 and thereafter indicating that there is no extension of the reproductive life span.

Expression of dppScer\UAS.cNa under the control of Scer\GAL4ey.PH leads to a reduction in the size of the eye disc, with the retinal field being enlarged at the expense of the rest of the disc. The antennal disc is broader and is partially duplicated. Adults of this genotype have heads that are significantly reduced in size, with misplaced, duplicated antennae and maxillary palps. Eyes are reduced in size and protrude from the head. The dorsal head is reduced in size but retains much of its normal appearance.

When dppScer\UAS.cNa is expressed under the control of Scer\GAL4Act5C.PI in several inner germarial sheath cells of the adult ovary, many spectrosome-containing single germ cells accumulate. The IGS cells that are expressing dppScer\UAS.cNa under the control of Scer\GAL4Act5C.PI maintain their normal morphology.

Overexpression of dppScer\UAS.cNa in somatic cells surrounding the female stem cell niche, driven by Scer\GAL4C587, causes germline stem cells (GSCs) to divide and accumulate, so that the number of GSCs vastly outnumbers those found in wild-type ovarioles. As with wild-type GSCs, GSCs expressing dppScer\UAS.cNa under the control of Scer\GAL4C587, delay cytokinesis. However, these GSCs lack the associated cap cells seen in wild type. Additionally, the new fusome of dppScer\UAS.cNa-expressing GSCs grows to the same size as the old fusome and do not stretch out into the "exclamation point" configuration characteristic of GSCs in contact with cap cells.

Expression of dppScer\UAS.cNa in inner sheath cells under the control of Scer\GAL4C587 blocks germ cell differentiation and results in the formation of germline stem cell-like tumours, leading to female sterility.

When dppScer\UAS.cNa is driven by Scer\GAL4C587 germ-line stem cell tumours are created in ovarioles.

In the testes of dppScer\UAS.cNa; Scer\GAL4nos.UTR.T:Hsim\VP16 adult males, the hub appears bigger with more cells, and there are slightly more single germ cells with a spectrosome around the hub cells. These testes also contain spermatogonial clusters with more than 16 germ cells.

dppScer\UAS.cNa; Scer\GAL4C587 germaria are filled with single germ cells, each with a single spectrosome. After 3 successive days of 2 hour heat-shock treatment, the anterior half of the germaria in dppScer\UAS.cNa; Scer\GAL4hs.PB adult females is filled with single-spectrosome containing germ cells.

Females expressing dppScer\UAS.cNa under the control of Scer\GAL4C587 have an increased number of primordial germ cells (PGCs) per gonad, averaging 245 +/- 92 per gonad compared to 136 +/- 26 in the wild type. 20.5 +/- of the PGCs are BrdU-positive in the mutant gonad compared to 15.8 +/- 2.0 % in the wild type. The mitotic activity of anterior PGCs is decreased in comparison to wild type, while the posterior PGCs are more mitotically active than wild type.

Wing discs expressing dppScer\UAS.cNa under the control of Scer\GAL4C-765 show significant overgrowth.

Eye discs expressing dppScer\UAS.cNa under the control of Scer\GAL4arm.PS appear to have a normal pattern of neural differentiation.

When expressed via Scer\GAL4hs.PB plus daily heat shock for 3 days germaria become tumorous. Large germline cells filling germarium regions 1 and 2a contain spectrosomes but show no evidence of cyst formation. Regions 2b and 3 show 16-cell cysts that probably derive from differentiated cystoblasts, or cysts that formed before the first heat shock. After 4-5 days of heat shock single germ line cells fill the germarium and 16 cell cysts are rarely detected. Germline cysts start to form in the most posterior part of the tumor 2 days after returning flies to normal temperatures. Many 16 cell cysts are seen by 4 days after the downshift, and must derive from tumor cells. Not all cysts contain 16 cells.

When expression is driven by Scer\GAL4C-765, even small clones induce significant over-proliferation. The large majority of cells in the outgrowth are wild-type and do not express dpp themselves.

Co-expression of dppScer\UAS.cNa with enokGD4037 and enokHM05195 under the control of Scer\GAL4bab1-Gal4 and Scer\GAL80ts.αTub84B is unable to attenuate the decline in germline stem cell numbers.

The Df(2L)s1402 deficiency can rescue the sterility of females that express dppScer\UAS.cNa under the control of Scer\GAL4C587. When dppScer\UAS.cNa is expressed under the control of Scer\GAL4C587 in a pelo1/+ background, 64% of ovarioles no longer show the stem cell tumour phenotype seen when dppScer\UAS.cNa is expressed in a wild-type background. These ovarioles contain differentiated germline cysts, developing egg chambers and mature eggs. When dppScer\UAS.cNa is expressed in a pelo1 homozygous background, the tumour phenotype is further suppressed.

When bamhs.PO is driven by heat shock in dppScer\UAS.cNa, Scer\GAL4C587 adults, cysts break down by a process of ring canal closure and fusome scission, de-differentiating. The multiple germ-line stem cells in each ovariole immediately and synchronously begin to differentiate after heat shock and soon generate 4-cell and 8-cell cysts. There are no single cells or cell pairs remaining at 20 h after heat shock. however by 30 h after heat shock (AHS) the cysts stop growing and start to beak down. By 40 h AHS closed ring canals and ring cell remnants are common. By 50 h AHS almost all germ cells are single.

Scer\GAL469B-mediated expression creates dorsalised embryos that undergo normal dorsal closure. Scer\GAL469B-mediated expression in Df(2L)flp147E embryos causes a partially dorsalised phenotype and partial rescue of the dorsal closure phenotype.

Expression of dppScer\UAS.cNa under the control of Scer\GAL4dpp.s-hc at 29oC causes an almost complete rescue of the head capsule defects of dppTgR46.1/Df(2L)DTD2, dppSal20 mutants. Expression at 27oC is less effective and causes a 55% reduction in penetrance while expression at 25oC results in a 22% reduction in penetrance.
