Detailed Description

Females carrying homozygous follicle cell clones can produce round eggs.

When somatic clones are made in the developing eye, man differentiated R-cell precursor nuclei are localised abnormally at the basal region. This appears to reflect a failure for R-cell precursor nuclei to migrate along the apical-basal axis. About 60% of nuclei exhibit this phenotype. When the size of mutant clone patches exceeds 40 ommatidia, a furrow like structure is often formed within the clone. This was always positioned in parallel to the morphogenetic furrow.

Approximately 30% of msn102 homozygous embryos shows a 'dorsal open' phenotype.

Many R1-R6 growth cones fail to stop at the lamina layer in msn03349/msn102 larvae.

Somatic clones in the eye leads to pathfinding defects in the photoreceptor axons. While photoreceptor axons derived from clones extend normally into the brain, the R1 to R6 axons fail to elaborate a smooth retinotopic array in the lamina. Many R1 ro R6 axons terminate prematurely giving rise to an uneven lamina neuropil. Surprisingly the growth cones that innervate the medulla appear to expand normally, although their projections are somewhat disorganised. msnP656A,P659A.Scer\UAS when driven by Scer\GAL469B in a msn102 background produces third instar larvae with severe defects in axonal targeting. On leaving the optic stalk axons from photoreceptors form very large axonal bundles and fail to defasciculate in the region of either the lamina or the medulla.

In msn102 somatic clones in the eye ommatidia often have malformed or missing photoreceptors. Those ommatidia that have the normal complement of photoreceptors are often misrotated and display the wrong chiral form or are symmetrical. In mosaic photoreceptor R3/R4 pairs, the msn+ photoreceptor has a strong preference for adopting the R3 fate. This can often lead to chirality inversions where the msn+ R4 precursor adopts the R3 position displacing the msn102 R3 precursor. The mosaic analysis also showed that for correct 90o rotation there is no bias for any particular photoreceptor.

Homozygous clones in the developing eye result in defects in photoreceptor cell innervation; the lamina appears uneven and abnormal, large bundles of axons are seen in the medulla. R1-R6 photoreceptor cell axons from mutant clones pass over the lamina and terminate abnormally in the medulla.

Homozygous or msn102/msn172 embryos show defects in dorsal closure.

Clones in the eye are externally rough. They contain elongated rhabdomeres and some ommatidia also contain missing or an extra photoreceptor.

msn102 does not protect Df(1)su(s)R194/+ clones in the eye; Df(1)su(s)R194/+ ; msn102 clones are not recovered in the adult eye in animals with mosaic eyes containing two genotypes of cells with respect to RpL36; cells which are Df(1)su(s)R194/+ and cells in which the haplo-insufficiency of Df(1)su(s)R194/+ for RpL36 has been rescued by RpL36+t4 (in a wild-type background the Df(1)su(s)R194/+ clones are eliminated by cell competition and are not seen in the adult eye in these animals). Also, the proportion of Df(1)su(s)R194/+ cells in the larval eye disc is not increased by the presence of msn102.

Introduction of a single copy of stck18 into the homozygous msn102 background reduces the incidence of a 'dorsal open' phenotype from approximately 30% to approximately 20%.

Homozygosity for icsBG02577 suppresses the msn102 'dorsal open' phenotype, reducing its incidence from approximately 30% to approximately 2%.

The photoreceptor projection phenotype caused by expression of bifScer\UAS.cRa under the control of Scer\GAL4GMR.PF is not modified by msn102.

msn102, suppresses the ommatidial polarity defects seen in dshhs.sev.B flies grown at 29oC. About 85% of ommatidia are in their correct polarity compared to 55% in dshhs.sev.B alone.

msn102/+ in double heterozygous combination with either bsk1/+, bsk2/+, bskflp147E/+, hep1/+ or hepr75/+ produces embryos with a dorsal open phenotype. The dorsal open phenotype of msn102/msn172 embryos is partially rescued by JraAsp.hs.sev expressed using heat shock or by tkvQ253D.Scer\UAS.cNb expressed under the control of Scer\GAL469B.
