tshScer\UAS.cGa under the control of Scer\GAL4GMR.PF results in roughening of the posterior portion of the adult eye as a result of nonspecific loss of photoreceptor cells within each ommatidium.

tshScer\UAS.cGa under the control of Scer\GAL4dpp.PS induces ectopic eye formation in the third-instar antennal imaginal disc and adult. The antennal discs show an increase in size compared to wildtype and have an abnormal profile.

tshScer\UAS.cGa under the control of Scer\GAL4cb26 induces ectopic eye formation in the third-instar antennal imaginal disc and adult. The antennal discs show an increase in size compared to wildtype and have an abnormal profile.

tshScer\UAS.cGa under the control of Scer\GAL4cb41 induces ectopic eye formation in the third-instar antennal imaginal disc and adult. The antennal discs show an increase in size compared to wildtype and have an abnormal profile.

Clones expressing tshScer\UAS.cGa, driven by Scer\GAL4αTub84B.PZ, in the eye disc margin or peripodial epithelium induce the overproliferation and columnarization of cells. Some of these clones show a sorting behaviour, with tshdsRNA.Scer\UAS-expressing cells arranging themselves as hollow sacs with their apical sides pointing inwards. Transient tshScer\UAS.cGa expression, driven by Scer\GAL4hs.PB following a heat shock to L2 larvae, results in the development of distinct ectopic clusters of photoreceptor cells in the peripodial epithelium. Such ectopic photoreceptors are seen when tshScer\UAS.cGa is expressed in the margin and the peripodial epithelium of the L3 disc by Scer\GAL4Bx-MS1096 or Scer\GAL4MD705. As adults these flies exhibit ectopic eyes. The Scer\GAL4Bx-MS1096>tshScer\UAS.cGa adults have ventral ectopic eyes are induced that replace ventral head structures and also show small dorsal patches of photoreceptors on the ventral head, which are normally accompanied by a bristle.

Ectopic expression of tshScer\UAS.cGa in the epidermis at 25oC, driven by Scer\GAL469B, causes a homeotic labial to T1 segment transformation, which is visible due to the formation of a denticle T1 "beard" in the labial segment. The maxillary and antennal segments also take on a trunk-like identity, shown by the development of ectopic denticles. Head involution is defective in these mutants and much of the head skeleton is missing. Additionally, these mutants experience a loss of denticles on the trunk.

Changes in cuticle pattern indicate that in tshScer\UAS.cGa; Scer\GAL469B embryos, the labial segment is transformed to T1 and the antennal and maxillary head segments are transformed towards a trunk identity in that they develop denticles and naked cuticle. tshScer\UAS.cGa; Scer\GAL4dpp.blk1 flies have an ectopic eye ventral to each of their eyes, whereas in tshScer\UAS.cGa; Scer\GAL4ey.PH flies the eyes are deformed.

When tshScer\UAS.cGa is driven by Scer\GAL469B, completely suppresses the formation of denticles.

Clones of cells in the eye disc expressing tshScer\UAS.cGa under the control of Scer\GAL4αTub84B.PZ grow poorly in the middle of the disc but can induce large overgrowths at the edge of the disc.

Pharate adults expressing tshScer\UAS.cGa under the control of Scer\GAL4dpp.blk1 show splitting of the endogenous eye in 8% of cases and sometimes have an ectopic eye at the base of the antenna. 42% of animals expressing tshScer\UAS.cGa under the control of Scer\GAL4dpp.blk1 arrest and die at the white pupal stage. These animals are about 50% larger than normal. These animals have an extended larval period (which is about 2-3 days longer than normal) and their eye-antennal discs show both overgrowth and distorted morphology. When two copies of tshScer\UAS.cGa are expressed under the control of Scer\GAL4dpp.blk1, the ventral half of the endogenous eye is missing in all flies. Expression of tshScer\UAS.cGa under the control of Scer\GAL4ey.PH results in complete suppression of eye development in 97% of cases. Expression of tshScer\UAS.cGa under the control of Scer\GAL4bi-omb-Gal4 results in absence of the ventral eye in pharate adults and overgrowth of the dorsal eye. The eye disc is enlarged in these animals. Expression of tshScer\UAS.cGa in clones under the control of Scer\GAL4Act5C.PI can suppress photoreceptor development in the eye disc, but only along the ventral margin. Clones in the dorsal margin of the eye disc result in overgrowth. Clones in the internal part of the eye, away from the margins, do not affect eye fate. The frequency of dorsal clones is higher than that of ventral clones. Adults in which clones expressing tshScer\UAS.cGa under the control of Scer\GAL4Act5C.PI have been induced show suppression of the ventral eye or overgrowth of the dorsal eye. Expression of tshScer\UAS.cGa under the control of Scer\GAL4dpp.blk1 or Scer\GAL4bi-omb-Gal4 results in splitting of the wing pouch. Expression of tshScer\UAS.cGa in clones under the control of Scer\GAL4Act5C.PI results in duplications of antennal segments distal to AN2. Clones in the anteroproximodorsal region of the antennal disc cause ectopic eye formation.

When tshScer\UAS.cGa is driven by Scer\GAL448Y no larvae hatch when reared at 25oC. When reared at 20oC a few escaper larvae are found. In these animals no copper cells are seen in the larval gut.

Expression of the P{UAS-tsh.G}20 insertion under the control of Scer\GAL469B does not affect denticle formation at 25oC.

Somatic clones ectopically expressing tshScer\UAS.cGa under the control of Scer\GAL4Act5C.PP or Scer\GAL4arm.T:Hsim\VP16 in the coxa and trochanter are wild-type. No clones are seen in ventral positions in the leg, however, vesicles consisting of bristleless cuticle are formed. Vesicles of cuticle are also formed in the lumen of the leg resulting in a reduction of femur length and a perturbation at the boundary region in any dorsal-ventral position. When clones are induced in third instar larvae, outgrowths are seen especially in dorsal positions. In distal parts of the femur, clones are observed within which the bristles lacked bracts. After a longer clone induction time, clones located in ventral positions are always abnormal, producing fusion between femur and tibia. Somatic clones ectopically expressing tshScer\UAS.cGa under the control of Scer\GAL4Act5C.PP or Scer\GAL4arm.T:Hsim\VP16 in the leg disc. two or three large clones per disc are seen in the endogenous tsh domain, mostly isolated cells are detected in the distal domain. these cells often sort out from their distal neighbors towards the lumen but remain attached to the epithelium of the disc. When induction time is increased, groups of tsh positive cells extend into the distal domain.

Expression of tshScer\UAS.cGa under the control of Scer\GAL469B results in embryos in which the head segments have trunk identity. These embryos have naked cuticle with almost no ventral denticle belts. The prothoracic beard is not present. Overexpression of tshScer\UAS.cGa in embryos using Scer\GAL4hs.PB between stages 9 and 10 results in a naked cuticle phenotype.

When expression is driven by Scer\GAL469B the ectopic sclerites of spenpoc361 are completely suppressed.

The lawn of denticles phenotype of pan3 or wgl-17 embryos is replaced by naked cuticle if the embryos are also expressing tshScer\UAS.cGa under the control of Scer\GAL469B. The lawn of denticles phenotype of arm2 embryos derived from arm2 female germ line clones is partly suppressed if the embryos are also expressing tshScer\UAS.cGa under the control of Scer\GAL469B.
