Ectopic expression of svScer\UAS.cKa under the control of Scer\GAL4sev.EP causes 26% of ommatidia to form one additional cone cell, an increase that is concomitant with a reduction in R7 photoreceptors. Compared to wild-type eyes, there is no significant change in primary pigment cell number in eyes expressing svScer\UAS.cKa via Scer\GAL4sev.EP.

Expression of svScer\UAS.cKa under the control of Scer\GAL4neur-GAL4-A101 results in external sensory organs with two (9%) and three (1%) hair cells as well as the normal one hair (90%).

Expression of svScer\UAS.cKa under the control of Scer\GAL4neur-GAL4-A101 in clones in the notum results in external sensory organs with multiple hair shafts, some with misshapen external cells and in some cases loss of external cells.

Pharate adults expressing svScer\UAS.cKa under the control of Scer\GAL4ey.PH not only lack eyes, but often have no head except for the proboscis, while the thorax and abdomen are wild type. The headless phenotype is fully penetrant but shows variable expressivity. Four classes of phenotype are seen; class I pharate adults lack all head structures derived from the eye-antennal discs, class II consists of eyeless flies which lack most head structures and both antennae, class III consists of eyeless animals with large parts of the head missing but one or both antennae present and class IV consists of animals in which most of the head and one or both eyes of reduced size are present.

Flies carrying svScer\UAS.cKa and Scer\GAL4hs.PB are normal when raised at 25oC. A 15 minute heat shock at 37oC during the pupal stage results in lethality, usually well before the pharate adult stage. A small fraction of the pupae develop to the pharate adult stage, and they all have some bristles on the notum and head that have extra shaft structures and lack sockets. Most commonly two smaller shafts are seen flanking a more normal-sized central shaft. Approximately 1/3 of the notal microchaetae are affected, and the orbital bristles on the head consistently show this phenotype. Flies expressing svScer\UAS.cKa under the control of Scer\GAL4Bx-MS1096 die as pharate adults with grossly malformed wings and bristle abnormalities. Bristles with extra shaft structures and lacking sockets are seen in particular regions of the notum and head, commonly having two smaller shafts flanking a more normal-sized central shaft. The microchaetae in the rows nearest the dorsocentral bristles consistently show this phenotype.

Co-expression of both prosL.Scer\UAS and svScer\UAS.cKa consistently converts one extra cell into a cone cell. In contrast, no change in photoreceptor cell or primary pigment cell number is observed.

Co-expression of svScer\UAS.cKa and prosdsRNA.Scer\UAS under the control of Scer\GAL4sev.EP significantly increases the transformation of R7 photoreceptors into cone cells, with the majority of ommatidia forming five cone cells and lacking R7s. Mild eye roughening is also observed.

Expression of svScer\UAS.cKa under the control of Scer\GAL4neur-GAL4-A101 in homozygous seqU5 clones results in external sensory organs with 1 hair (89%) or 2 hairs (11%).

Expression of both svScer\UAS.cKa and hamScer\UAS.cMa under the control of Scer\GAL4neur-GAL4-A101 in clones in the notum results in the loss of external cells in almost 100% of external sensory organs in the clone.
