Expression of pksple.Scer\UAS under the control of Scer\GAL4A51 or Scer\GAL4elav-C155 does not lead to bang sensitivity.

Expression of pksple.Scer\UAS under the control of Scer\GAL4Toll-6-D42 results in a large fraction of motor neuron microtubules showing reversed polarity, but no significant difference in plus-end polymerization as compared to controls

Overexpression of pksple.Scer\UAS using Scer\GAL4hs.2sev induces ommatidial chirality and rotational defects in the eye and wing hair orientation defects between wing veins L1 and L4.

Scer\GAL4C-765-driven overexpression of pksple.Scer\UAS results in altered wing membrane ridge orientation and reversal (disto-proximal) hair polarity across most of the wing. Wing membrane ridges on Scer\GAL4C-765/pksple.Scer\UAS wings appear rotated anti-clockwise compared to wild-type, with the exception of the proximal-distal ridges immediately anterior to the compartment boundary. Hair polarity and ridge orientation are approximately orthogonal in both the anterior and posterior of Scer\GAL4C-765/pksple.Scer\UAS wings. This orthogonal relationship between hairs and wing membrane ridges is normally only seen in the anterior of the wing.

All Scer\GAL4hs.PB; pksple.Scer\UAS pupae heat-shocked between 0 and 28 h after puparium formation (a.p.f.) show adult wing phenotypes.

Induction of Scer\GAL4hs.PB-driven pksple.Scer\UAS from 0 to 7 h a.p.f. results in an increasing area of the wings displaying altered hair polarity and wing membrane ridge orientation, however, these phenotypes were variable and difficult to interpret.

Induction of Scer\GAL4hs.PB-driven pksple.Scer\UAS between 8 and 10 h a.p.f. alters wing membrane ridge orientation in both the anterior and posterior regions of wings, with the exception of some distal regions which retain wild-type ridge orientation. Hair polarity and wing membrane ridge orientation in both the anterior and posterior regions of the Scer\GAL4hs.PB/pksple.Scer\UAS wings are approximately orthogonal, even in those regions where wing membrane ridges still have wild-type orientation.

Wing membrane ridge phenotypes are evident up until induction of Scer\GAL4hs.PB-driven pksple.Scer\UAS at 15 h a.p.f., although they became increasingly confined to proximal wing regions with later induction. Wing membrane ridge orientation is primarily proximal-to-distal in both the anterior and posterior regions of the Scer\GAL4hs.PB/pksple.Scer\UAS wings and hair polarity and wing membrane ridge orientation appear approximately orthogonal across the entire wing.

Induction of Scer\GAL4hs.PB-driven pksple.Scer\UAS after 15 h a.p.f. results in primarily wild-type (proximal-to-distal) wing membrane ridge orientation in the posterior of the wing. Anterior wing membrane ridge orientation is predominantly proximal-to-distal compared to anterior-to-posterior in wild-type. Hair polarity and wing membrane ridge orientation are approximately orthogonal in both the anterior and posterior regions of wings, resulting in a strong posterior component to hair polarity in the anterior of the wing and a strong anterior component to hair polarity in the posterior of the wing

Expression of pksple.Scer\UAS under the control of Scer\GAL4Act in the wing results in wing hairs pointing proximally and anteriorly.

In the abdomens of pksple.Scer\UAS; Scer\GAL4αTub84B.PL flies, the planar polarity of the entire posterior compartment is reversed. This reversal extends slightly in to the anterior end of the anterior compartment, which is otherwise largely unaffected. This reversal phenotype is also seen in pksple.Scer\UAS; Scer\GAL4en-e16E and pksple.Scer\UAS; Scer\GAL4hh-Gal4 flies. This phenotype is not presenting pksple.Scer\UAS; Scer\GAL4ci.PH animals which have only some localised disturbance of planar polarity at the fronts of both anterior and posterior compartments in the tergite.

In the posterior compartment of the abdomen of pksple.Scer\UAS; Scer\GAL80αTub84B.PL; Scer\GAL4αTub84B.PL flies, somatic clones lacking Scer\GAL80αTub84B.PL have reversed tissue polarity in all cells apart from some of the most anterior in each clone. Some wild-type cells immediately behind these clones usually have reversed polarity. Clones in the anterior compartment have normal polarity, apart from those at the posterior of the compartment, which have a zone of reversed polarity cells in posterior compartment cells posterior to them.

When pksple.Scer\UAS is driven by Scer\GAL4hs.2sev typical planar cell polarity defects are seen in the eye: symmetric ommatidia and chirality and rotation defects. About 27% of ommatidia exhibiting abnormal polarity.

When expression is driven by Scer\GAL4Act5C.PI wing hairs point proximally throughout the wing.

When pkScer\UAS.cGa is driven by Scer\GAL4ptc-559.1, the wing hairs in the region between wing vein L3 and L4, distal to the anterior crossvein point towards the midline of the wing, unlike wild-type wings, where the wing hairs point distally.

Over expression of pksple.Scer\UAS when driven by Scer\GAL4da.G32 or Scer\GAL4C-765 gives a pk phenotype in the wing triple row bristles; the tarsi however remain wild-type. Also, when driven by Scer\GAL4da.G32, swirls are seen in the wing hair orientation with different patterns from wing to wing.
