Expression of linScer\UAS.cHa under the control of Scer\GAL4Ubx-lac1-Gal4 results in flies lacking both heminota.

Expression of linScer\UAS.cHa under the control of Scer\GAL4Act5C.PU in large clones induced during the first larval instar results in flies lacking a heminotum.

Expression of linScer\UAS.cHa under the control of Scer\GAL4klu-G410 results in a reduction in the extent of the prescutum and eliminates lateral microchaetae in the ventral prescutum.

Larvae expressing linScer\UAS.cHa under the control of Scer\GAL4dpp.blk1 show antennal duplication in both antennal discs and pharate adults, or formation of ventral eyelets in the head capsule.

The wing discs have a spherical morphology in animals expressing linScer\UAS.cHa under the control of Scer\GAL4Ubx-lac1-Gal4. The thin peripodial epithelium is replaced with a mirror image duplication of the thickened, pseudostratified columnar epithelium that forms the disc proper. The mutant discs are approximately 10-30% the size of wild-type wing discs. The number of actively dividing cells in the mutant discs is similar to wild type, but the number of apoptotic cells is elevated in the mutant discs. The mutant discs survive beyond the larval stages and elongated during metamorphosis, but fail to contribute to adult structures.

Large clones in the disc proper of the wing disc that are expressing linScer\UAS.cHa under the control of Scer\GAL4Act5C have no effect on wing development.

Large clones in the peripodial epithelium of the wing disc that are expressing linScer\UAS.cHa under the control of Scer\GAL4Act5C completely transform the peripodial epithelium into disc proper tissue, resulting in a mirror image duplication of the pseudostratified columnar epithelium. These discs are spherical and smaller than normal.

Smaller clones in the peripodial epithelium of the wing disc that are expressing linScer\UAS.cHa under the control of Scer\GAL4Act5C result in the formation of secondary wing fields in the discs. The presumptive notum appears intact in some of these discs, but is reduced or missing in others. The clones that produce this phenotype adopt disc proper fate and occupy approximately 26% of the surface area of the entire wing disc.

Peripodial epithelium cells in the wing disc that are expressing linScer\UAS.cHa under the control of Scer\GAL4ptc-559.1 adopt disc proper fate and permit the induction of a secondary wing field in the disc.

Clones at the margin in the eye disc that are expressing linScer\UAS.cHa under the control of Scer\GAL4Act5C.PP result in a failure of retinal initiation.

The 1o-3o fates of the dorsal epidermis are replaced with the 4o fate in embryos expressing linScer\UAS.cHa under the control of Scer\GAL4ptc-559.1.

Embryos expressing linScer\UAS.cHa under the control of Scer\GAL4byn-Gal4 have a short, wide hindgut, consisting of an undulating layer of columnar cells with a duplication of the boundary cell rows.

Expression of linScer\UAS.cHa under the control of Scer\GAL4arm.PS results in replacement of 1o-3o fates in the dorsal cuticle (dorsal denticles, smooth cuticle and thick hairs respectively) with 4o fates (fine hairs). Expression of linScer\UAS.cHa under the control of Scer\GAL4ptc-559.1 is occasionally not sufficient to specify the 4o dorsal cuticle fate (fine hairs) in place of the 2o fate (smooth cuticle). Expression of linScer\UAS.cHa under the control of Scer\GAL4en-e16E in linG2/lin2 embryos transforms all en-expressing cells to the 4o fate (fine hairs).

The 4o dorsal cuticle fate is restored in embryos expressing panΔN.Scer\UAS under the control of Scer\GAL4ptc-559.1 if they are also coexpressing linScer\UAS.cHa; 4o type cells differentiate in place of the 3o cells, in cells flanking the en expression domain. The ectopic 1o cell row is not rescued. Anterior to this ectopic row, smooth cuticle is replaced with the 4o cell fate.

Expression of linScer\UAS.cHa under the control of Scer\GAL4ptc-559.1 rescues ventral and dorsal wg-dependent cell fates in lin2/linG2 embryos. Anterior ventral denticle rows are restored and excess row 2/3 denticles are produced. The excess denticles posterior to the belt that are seen in lin2/linG2 embryos are not restored to smooth cuticle. Expression of linScer\UAS.cHa under the control of Scer\GAL4wg.PM in linG2/lin2 embryos restores the 4o fate, but only in the portion of the 4o field that expresses wg.
