Embryos from krzS047819 maternal germline clones show delayed ventral furrow formation and large ventral holes. The same is observed in embryos from krzS047819 homozygous mothers rescued to adulthood by krzVL-A.Tag:SBP.

krzd1/krzS047819 mutant animals survive until the third larval instar, where they become immobile and flaccid and form melanotic tumors. The imaginal discs of these mutant larvae are very reduced in size and display increased cell death.

Expression of krzScer\UAS.cMa in the central nervous system under the control of Scer\GAL4wor.PA rescues the lethality of krzd1/krzS047819. The surviving flies are sterile and display slightly folded wings. Wing discs in rescued larvae have a normal appearance.

49% of krzS047819 mutant cuticles display a large hole either at the anterior or posterior region of the embryo, 31% of mutant embryos display two holes, and 17% of the cuticles display twisting along the anterior-posterior axis. Mutants display gastrulation defects and embryonic lethality.

The embryonic nervous system is disrupted in stage 15 krzS047819 embryos, and animals display holes in the nerve cord, or a twisted nerve cord phenotype.

Homozygous adults which have been rescued from lethality by expression of krzScer\UAS.cRa under the control of Scer\GAL4hs.PB during development (using twice daily heat shocks) die at a faster rate than heterozygous siblings or wild-type controls on medium containing no added NaCl. On medium containing 0.4M NaCl, homozygous adults die at a much faster rate than heterozygous siblings or wild-type controls.

krzS047819 homozygous adults which have been rescued from lethality by expression of krzScer\UAS.cRa under the control of Scer\GAL4hs.PB show a reduced initial activity phase (during the first minute) in an open field arena compared to control flies. The spontaneous locomotor activity is not significantly different from control flies from the second minute onwards (the assay was carried out for 10 minutes).

krzS047819 homozygous adults which have been rescued from lethality by expression of krzScer\UAS.cRa under the control of Scer\GAL4hs.PB and which are also expressing krzScer\UAS.cRa under the control of Scer\GAL4Or83b.T:Hsim\VP16 still show a reduced initial activity phase in an open field arena compared to control flies.

There are no significant differences in spontaneous activity between krzS047819 homozygous adults which have been rescued from lethality by expression of krzScer\UAS.cRa under the control of Scer\GAL4hs.PB and control flies measured in the dark over a period of days.

krzS047819 homozygous adults which have been rescued from lethality by expression of krzScer\UAS.cRa under the control of Scer\GAL4hs.PB fail to show a robust increase in activity after mechanical stimulation, in contrast to control flies.

The majority of homozygous krzS047819 mutants die as embryos, and less than 25% reach the third instar larval stage

Homozygotes die throughout development. Most animals fail to hatch and none of the surviving larvae live to puparium formation. Prior to death the mutant larvae become immobile and flaccid. Third instar larvae develop melanotic tumours, which form throughout the body cavity but most frequently collect in the posterior of the larva. Tumours are frequently found floating free in the haemocoel. Large numbers of lamellocytes are found circulating through the haemocoel of mutant third instar larvae (these cells typically form in wild-type pupae). The fat bodies of mutant larvae shortly after the second moult have a rough appearance compared to those of wild-type larvae. At approximately 18-20 hours after the second moult the fat bodies of surviving mutant larvae begin to dissociate into individual cells (the wild-type fat body does not normally dissociate until after pupariation). Melanotic tumours begin to form on the surface of the mutant fat bodies shortly after the second moult.

Expression of krzScer\UAS.T:Zzzz\GS-TAP restores the regular ommatidial pattern and size of the eye in animals expressing rlSem.S.Scer\UAS under the control of Scer\GAL4da.G32 in a krzS047819/+ mutant background.

The survival rate of animals expressing rlSem.S.Scer\UAS under the control of Scer\GAL4da.G32 drops to 8% in a krzS047819/+ mutant background.

The survival rate of animals expressing rlSem.S.Scer\UAS under the control of Scer\GAL4da.G32 in a krzS047819/+ mutant background increases to 38% when krzScer\UAS.T:Zzzz\GS-TAP is co-expressed.

The notching and vein thickening phenotypes seen in dxS1M hemizygous males are enhanced by krzS047819/+. Survival of NAx-9/NAx-E2 dxENU/+ flies to adulthood is significantly decreased by krzS047819/+.
