mask10.22/Df(3R)BSC317 wandering third instar larvae have more autophagosomes in somatic muscles.

mask10.22/Df(3R)BSC317 mutant third instar larvae display reduced connectivity of the mitochondrial network as well as defects in the subcellular distribution of mitochondria in a body wall muscle.

Homozygous clones in the eye-antennal disc are significantly smaller than wild-type twin spot clones. The mutant cells do not appear to be significantly smaller than wild-type cells. The mutant cells show an increase in apoptosis compared to the wild-type cells. Some cells within clones in the third larval instar eye disc begin photoreceptor differentiation, but the ommatidia within clones contain less than the normal number of photoreceptor cells. Adult flies show loss of outer and inner photoreceptor cells within many clonal ommatidia. Analysis of mosaic ommatidia containing both wild-type and homozygous cells indicates that there is a partial requirement for mask function for the differentiation of R1-R8 photoreceptor cells. The R7 or R6 cell is never homozygous in mosaic ommatidia and the number of homozygous R1 cells in mosaic ommatidia is significantly reduced. Large homozygous clones in the eye (generated using the "ey-flp" method) can proliferate and form much of the eye. However, many photoreceptors, including the majority of R7 cells, fail to differentiate. Eggs derived from females containing homozygous follicle cells have fused or closely apposed dorsal appendages.

The indentations on the thorax of adult males hemizygous for Pink1B9 as well as their reduced ability to fly can be partially suppressed, in a dose-dependent manner, by combination with mask10.22/+ or mask10.22/maskEY13048 alleles, which leads to significantly lower penetrance of the thoracic phenotype and increased flight ability compared to Pink1B9 mutants alone.

The disturbed morphology of both mitochondria and indirect flight muscles characteristic for adult Pink1B9 hemizygote males is substantially suppressed by combination with mask10.22/+ or mask10.22/maskEY13048 alleles.

The indentations on the thorax of adult park25/parkΔ21 transheterozygotes as well as their reduced ability to fly can be partially suppressed, in a dose-dependent manner, by combination with mask10.22/+ or mask10.22/maskEY13048 alleles, which leads to significantly lower penetrance of the thoracic phenotype and increased flight ability compared to park25/parkΔ21 mutants alone.

The disturbed morphology of both mitochondria and indirect flight muscles characteristic for adult park25/parkΔ21 transheterozygotes is substantially suppressed by combination with mask10.22/+ or mask10.22/maskEY13048 alleles.

The enlarged or aggregated mitochondria phenotype observed in the body wall muscles of park25/parkΔ21 transheterozygote third instar larvae is suppressed by combination with mask10.22/maskEY13048 alleles.

The enlarged or aggregated mitochondria phenotype observed in the body wall muscles of Pink1B9 hemizygous male third instar larvae is suppressed either by combination with mask10.22/maskEY13048 alleles or by ectopic expression of parkScer\UAS.cGa under the control of Scer\GAL4how-24B.

Enhances the cswCS.hs.2sev rough eye phenotype. mask10.22 dominantly suppresses the ectopic R7 phenotype caused by sevS11.T:Hsap\MYC; the double mutant flies have an average of 1.8 ectopic R7 cells per ommatidium. mask10.22 dominantly suppresses the ectopic R7 phenotype caused by csw::Src64Bhs.2sev; the double mutant flies have an average of 0.6 ectopic R7 cells per ommatidium. One copy of mask10.22 weakly suppresses the Ras85DV12.sev rough eye phenotype. One copy of mask10.22 weakly enhances the Ras85DN17.hs.2sev rough eye phenotype. The number of apoptotic cells in cswCS.hs.2sev third larval instar eye discs is increased, both anterior and posterior to the morphogenetic furrow if the flies are also carrying one copy of mask10.22. The rlD334N.hs.sev ectopic R7 cell phenotype is partially suppressed by mask10.22/+; the double mutant flies have an average of 1.2 ectopic R7 cells per ommatidium.

Comments

Expressing maskUAS.cZa under the control of Scer\GAL4how-24B rescues the over-accumulation of autophagosomes and increases the amount of autolysosomes in somatic muscles from mask10.22/Df(3R)BSC317 wandering third instar larvae.
