Description

Imprecise excision of P{lacZ}dallyP1 has lead to a deletion of -724 to +415 od dally (Where the A in the ATG start codon is designated as 1).

dally80 heterozygotes resist infection with wild-type group B streptococcus.

dally80 heterozygotes survive longer than wild-type flies when infected with ACP-GAG binding impaired group B streptococcus.

The ovarioles of homozygous females show loss of the germline stem cells (GSCs), with 45% of ovarioles containing no GSCs in the niche at day 3 after eclosion.

The ovarioles of dally06464/dally80 females show progressive loss of the germline stem cells (GSCs), with 59% of ovarioles containing no GSCs in the niche at day 10 after eclosion.

Wings of homozygous adults are smaller than normal, primarily due to a reduction in cell number rather than cell size.

dally80/Df(3L)Scf-R6 flies show a reduction in the number of chemosensory bristles at the anterior margin of the wing compared to wild-type flies. The L5 vein in the wings of these flies does not form properly and stops short between the posterior crossvein and the wing margin. These mutants also show a loss of the anterior dorsocentral bristles and the posterior supraalar bristles on the notum.

When dally80 mutant clones are made in the wing disc a loss of sensory organ precursors are seen. Mutant adults escapers exhibit reduced number of chemosensory bristles on the wing.

Somatic clones of dally80 homozygous cells in the wing lead to a partial loss of the L5 vein with a high penetrance, but no detectable defects in the central domain of wing blade.

Wing disc clones mutant for dally80 and dlpA187 often have incomplete or fragmented posterior crossveins. This effect is not cell-autonomous.

The commissural axon phenotype (failure to cross the midline) seen in embryos expressing Sema-1aScer\UAS.cYa under the control of Scer\GAL4P52 in a Sema-1ak13702 null background is significantly enhanced if the embryos are also heterozygous for dally80.

dally80 dlpA187 Df(2R)48 triple heterozygotes exhibit increased median survival time compared to controls when infected with group B Streptococcus (both wild-type and GAG-binding impaired mutants) and lower colony counts after infection.

dally80 Sulf1Δ1 double mutants exhibit a phenotype similar to dally80 single mutants (a reduced number of bristles on the wing margin).

The ability of dlp-HS.Scer\UAS expressed under the control of Scer\GAL4prd.RG1 to rescue naked cuticle patterning in dlpA187 embryos derived from dlpA187 female germline clones is not affected if the embryos are also mutant for dally80.

dally80 dlpA187 double heterozygotes resist infection with wild-type group B streptococcus (to a greater extent than single mutants).

dally80 dlpA187 Df(2R)48 triple heterozygotes resist infection with wild-type group B streptococcus (to a greater extent than single and double mutants).

dally80 dlpA187 Df(2R)48 triple heterozygous mutants survive longer than wild-type flies when infected with ACP-GAG binding impaired group B streptococcus.

dlpA187, dally80 mutants display ISNb bypass at a similar rate to wild-type embryos. The Larbypass ISNb phenotype is not enhanced in Larbypass; dlpA187, dally80 triple mutants.

When dally80/NotumI6 double mutant clones are made in the wing disc, reduced numbers of sensory organ precursors are seen in large clones.

Somatic clones of dally80; dlpA187 doubly homozygous cells in the wing lead to L3-L4 fusion, a phenotype not seen with single mutants clones.

One copy of dally80 partially suppresses the wing margin defects seen when Drer\wif1Scer\UAS.cAa is expressed under the control of Scer\GAL4nub-AC-62.

Complementation and Rescue Data
Comments

Expression of dallyScer\UAS.T:Avic\GFP under the control of Scer\GAL4en-GAL4-33 increases the size of both the posterior and anterior compartments of the wing in dally80 adults.

Expression of dallyΔgpi.Scer\UAS.T:Avic\GFP under the control of Scer\GAL4en-GAL4-33 increases the size of the anterior compartment but not the posterior compartment of the wing in dally80 adults.

Expression of dallyΔGAG.Scer\UAS under the control of Scer\GAL4A9 partially rescues the phenotypes of dally80/Df(3L)Scf-R6 mutants. The transgene substantially restores chemosensory bristle numbers in the wing margin, almost fully rescues the formation of wing vein L5, almost fully restores the formation of the anterior dorsocentral bristles on the notum without inducing the formation of extra ones for all mutants. However, it fails to restore formation of the posterior supraalar bristles.
