The expression of VGlutUAS.cDa under the control of Scer\GAL4VGlut-OK371 induces synaptic transmission defects in the larval muscle 6/7 NMJ: recordings conditions with extracellular Cl show an increase in mEPSP and mEPSC amplitudes, decreased quantal content, increased failure rate and no paired-pulse depression; recording conditions without extracellular Cl also increase in mEPSC and EPSC amplitudes and unaffected quantal content.

Larvae expressing VGlutUAS.cDa under the control of Scer\GAL4VGlut-OK371 have defective neuromuscular junction neurotransmission: increased mEJP amplitude and decreased quantal content, but unaffected EJP amplitudes.

The expression of VGlutScer\UAS.cDa under the control of Scer\GAL4VGlut-OK371 leads to larval neuromuscular junctions under 0.4mM extracellular Ca[2+] concentration conditions exhibiting a significant increase in miniature excitatory postsynaptic potentials and a reciprocal significant decrease in quantal contents, but no obvious changes in evoked excitatory postsynaptic potentials, as compared to controls. Under 0.2mM extracellular Ca[2+] concentration conditions, these neuromuscular junctions exhibit significant decreases in evoked excitatory postsynaptic potentials and quantal contents, as compared to controls.

Intracellular voltage recordings in muscles of animals overexpressing VGlutScer\UAS.cDa driven by Scer\GAL4VGlut.PD or Scer\GAL4BG380 reveal large spontaneous events in motoneurons.

Driving VGlutScer\UAS.cDa withe the strong Scer\GAL4BG380 driver results in death during larval stages.

Driving VGlutScer\UAS.cDa with Scer\GAL4VGlut.PD results in lethality within the first few weeks of adulthood.

Flies overexpressing VGlutScer\UAS.cDa driven by Scer\GAL4VGlut.PD are uncoordinated and cannot fly. VGlutScer\UAS.cDa-overexpressing flies display progressive motor defects manifesting in impaired negative geotaxis.

Expression of VGlutScer\UAS.cDa in post-mitotic neurons under the control of Scer\GAL4elav-C155 causes a significant increase in average mEPSP amplitude. There is also a homeostatic decrease in presynaptic quantal content in these mutants.

Males expressing VGlutScer\UAS.cDa under the control of Scer\GAL4αTub84B.PL show high levels of homosexual courtship, which includes all aspects of sexual behaviour (including singing, genital licking and attempted copulation). Occasionally, the males attempt copulation with inappropriate body reions, such as the head.

Males expressing VGlutScer\UAS.cDa under the control of Scer\GAL4NP0225 show a significant amount of homosexual behaviour, although less than seen when the Scer\GAL4αTub84B.PL driver is used.

Expression of VGlutScer\UAS.cDa under the control of Scer\GAL4Mef2.247 has no affect on the courtship behaviour of males.

Expression of VGlutScer\UAS.cDa under the control of Scer\GAL4BG380 results in an increase of about 60% in the mean mEJP amplitude recorded from the muscle of the larval neuromuscular junction compared to wild type. The amplitude of the entire population of mEJP events seems to be increased in size, with the median amplitude increasing by 35% compared to wild type. There is a small increase in mEJP frequency. Larvae expressing VGlutScer\UAS.cDa under the control of Scer\GAL4BG380 show no significant difference in the amplitude of evoked EJPs recorded from the muscle of the larval neuromuscular junction compared to wild type. Quantal content is decreased relative to wild type. Larvae expressing VGlutScer\UAS.cDa under the control of Scer\GAL4BG380 show an increase in synaptic vesicle size (assayed at the neuromuscular junction of the type Ib motorneuron that innervates muscles 6 and 7) compared to wild type; a 14% increase in mean vesicle outer diameter and a median increase in vesicle outer diameter of 10% is seen.

The increased miniature excitatory postsynaptic potentials and the reciprocal decreased quantal contents under 0.4mM extracellular Ca[2+] concentration conditions observed at the larval neuromuscular junctions of individuals expressing VGlutScer\UAS.cDa under the control of Scer\GAL4VGlut-OK371, however, are not significantly altered by KaiR1DMB01010 homozygosity.

Scer\GAL4elav-C155 overexpression of VGlutScer\UAS.cDa significantly decreases the number of satellite boutons seen in Mon1Δ181/Df(2L)Exel9062 third instar larvae. Scer\GAL4elav.PU overexpression of VGlutScer\UAS.cDa significantly decreases the number of satellite boutons seen in Mon1Δ181/Mon1Δ181 third instar larvae.

Expression of VGlutScer\UAS.cDa and gsb491.dsRNA.Scer\UAS in post-mitotic neurons under the control of Scer\GAL4elav-C155 causes a significant increase in average mEPSP amplitude. There is also a homeostatic decrease in presynaptic quantal content in these mutants.
