Expressing dsScer\UAS.cTa under the control of Scer\GAL4hh.PU results in smaller relative posterior wing compartment area, as compared to wild-type controls.

Expressing dsScer\UAS.cTa under the controls of Scer\GAL4en.PU results in smaller anterior-posterior cross-vein distance as compared to wild-type controls.

Expression of dsScer\UAS.cTa under the control of Scer\GAL4hh.PU in a ds38k/ds36D background leads to high lethality during early pupal stages, with only a few adult survivors that show severe wing defects, as compared to ds38k/ds36D controls.

Overexpression of dsScer\UAS.cTa under the control of Scer\GAL4Bx-MS1096 results in wing shape, posterior ridge and wing hair polarity phenotypes.

Overexpression of dsScer\UAS.cTa under the control of Scer\GAL4salm-459.2 does not affect wing hair polarity.

At 24 hours after puparium formation, the majority of microtubules in wing cells within the expression domain of animals co-expressing dsScer\UAS.cTa and dsEP-822 under the control of Scer\GAL4en-e16E in the posterior wing compartment align themselves along the proximodistal axis, as occurs in wild-type wing cells. Wing hair polarity in the posterior compartment is not disrupted in the mutant adults, but the adjacent anterior cells (up to 5-6 cells) misorient their hairs away from the anterior/posterior compartment boundary.

Generalized overexpression of dsScer\UAS.cTa under the control of Scer\GAL4arm.PS disrupts denticle belt row structure and disturbs denticle polarity, most strongly in the A compartment.

Overexpression of dsScer\UAS.cTa in the P compartment (under the control of Scer\GAL4en-e16E) depolarizes denticle belt row 2 while row 3 polarity is reversed to now point forwards. This effet on row 3 indicates that there has been a propagation of a polarity change to the next cell.

Scer\GAL4ptc-559.1-led expression of dsScer\UAS.cTa depolarized denticle belt row 2 in the third stage larva. Both rows 1 and 3 point up towards the neighboring cells of row 2 and, in both, their polarity is reversed from that of the wild-type. Rows 4-6 are unaffected. Row 0, as is normal, points forwards. These changes in polarity are not accompanied by any gross changes in denticle shape or size suggesting that they retain their proper identity.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing dsScer\UAS.cTa under the control of Scer\GAL4αTub84B.PL in a wild-type background can result in a reversal of cell polarity in wild-type cells posterior to the clone (hairs behind the clone point forwards). This phenotype is weak.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing dsScer\UAS.cTa under the control of Scer\GAL4αTub84B.PL in a dsunspecified background do not change the cell polarity of cells surrounding the clone.

S2 cells that express dsScer\UAS.cTa under the control of Scer\GAL4Act5C.PI, aggregate.

Expression of dsScer\UAS.cTa in the wing causes hairs to orient away from the region of misexpression when driven by Scer\GAL4salm-459.2, Scer\GAL4Dll-md23 or Scer\GAL4en-e16E.

Wings that express dsScer\UAS.cTa under the control of Scer\GAL4αTub84B.PL causes a foreshortening along the proximodistal axis of the wing blade as the anterior and posterior crossveins are abnormally close.

dsScer\UAS.cTa; Scer\GAL4Act5C transfected S2 cells do not aggregate. dsScer\UAS.cTa; Scer\GAL4salm-459.2 wings have minor planar polarity defects, with a small region proximal to the posterior crossvein being most severely effected, and a region distal to the anterior crossvein, which is truncated, being more mildly affected. dsScer\UAS.cTa; Scer\GAL4en-e16E also have only mild planar polarity, restricted to a region of the proximal wing immediately anterior to the anterior-posterior compartment boundary. dsScer\UAS.cTa; Scer\GAL4Dll-md23 wings have a more severe planar polarity phenotype, with the normal wing hair polarity largely restricted to the regions close to the anterior-posterior boundary apart from its distal tip. dsScer\UAS.cTa; Scer\GAL4αTub84B.PP wings, by contrast, have completely normal wing hair planar polarity. Experiments using Scer\GAL80ts.αTub84B indicate that the expression of ds using dsScer\UAS.cTa; Scer\GAL4salm-459.2 is only able to disrupt planar cell polarity in adult wings when it occurs in a window around late larval and early pupal stages (before 18 hours after puparium formation).

Expression of dsScer\UAS.cTa under the control of Scer\GAL4hh.PU fails to rescue the wing defects of ds38k/ds36D adults.

Expression of dsScer\UAS.cTa, under the control of Scer\GAL4αTub84B.PL, rescues the planar cell polarity defects of ds05142 wings in all areas except the most proximal regions of the wing blade. Expression driven by Scer\GAL4ds-gal4 partially rescues planar cell polarity in the proximal wing but does not fully rescue the loss of the crossveins.

Uniform dsScer\UAS.cTa expression using Scer\GAL4αTub84B.PP rescues the planar cell polarity defects of ds05142/ds05142 flies in the distal region between L3 and L4, but not in the proximal region just posterior to L1.
