The expression of ykiUAS.cHa under the control of Scer\GAL4ptc.PU, Scer\GAL4hh.PU and Scer\GAL4nub.PU leads to an expansion of the anterior-posterior boundary region of the wing disc, the posterior region of the wing disc and the full wing pouch, respectively.

The expression of ykiUAS.cHa from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] leads to the third instar larval wing disc dorsal compartment presenting a significant increase in size, but does not lead to polyploidy/aneuploidy or epithelial organization defects (i.e. cells show normal sub-cellular localization of Discs large protein), as compared to controls; these larvae eventually enter pupariation but die as pupae. Expression under the control of either Scer\GAL4hh.PU or Scer\GAL4e22c also leads to a larger third instar larval wing disc posterior compartment, as compared to controls.

Overexpression of ykiScer\UAS.cHa driven in the fat body of male flies by Scer\GAL4r4 (or driven by Scer\GAL4yolk in female fat bodies) significantly decreases survival of flies in response to Gram-positive bacteria (Staphylococcus aureus or Enterococcus faecalis) and a fungus (Aspergillus fumigatus), compared to controls; expression with Scer\GAL4r4 in males has no effect on survival in response to Gram-negative bacteria (Erwinia carotovora carotovora 15 or Escherichia coli). Bacterial load (measured as colony forming units 24hr post infection) of S. aureus (but not Erwinia carotovora carotovora 15) significantly increases in adult flies, compared to controls.

Third instar larvae expressing ykiScer\UAS.cHa under the control of Scer\GAL4RapGAP1-OK6 show normal number of satellite boutons on neuromuscular junctions relative to controls.

Expression of ykiScer\UAS.cHa under the control of Scer\GAL4C587 has no apparent germarium phenotype.

Expression of ykiScer\UAS.cHa under the control of Scer\GAL4en.PU in third instar larvae leads to increased area of the posterior compartment of the wing disc.

The size advantage of ykiScer\UAS.cHa clones and the induction of apoptosis in wild-type cells indicates cell competition.

Adherens junctions of wing disc cells expressing ykiScer\UAS.cHa under the control of Scer\GAL4en-e16E are 1.78 times wider than those of wild-type control cells in the same disc.

Expression of ykiScer\UAS.cHa under the control of Scer\GAL4GMR.PF results in overgrown eyes.

Expression of ykiScer\UAS.cHa in the germline, under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 or Scer\GAL4bam.T:Hsim\VP16, does not result in germline over-proliferation in testes or ovarioles.

Expression of ykiScer\UAS.cHa under the control of Scer\GAL4GMR.PF results in increased eye size compared to controls.

Overexpression

of ykiScer\UAS.cHa in the developing eye, driven by Scer\GAL4GMR.PF, results in the generation of excess interommatidial pigment and bristle cells. Wild-type retinae have 1.67 cells in regions corresponding to one side of an

ommatidial hexagon between two vertices, while Scer\GAL4GMR.PF>ykiScer\UAS.cHa retinae have 5.9 cells in the same region.

Clones in the wing disc that express ykiScer\UAS.cHa under the control of Scer\GAL4tub grow much larger than wild-type controls. These clones cause the epithelium to bulge apically, due to a change in cell morphology in which nuclei move to a more apical position.

Expression of yki in the wing disc, under the control of Scer\GAL4hh-Gal4 results in an overgrowth of the posterior compartment.

Clones in the eye imaginal disc expressing ykiScer\UAS.cHa under the control of Scer\GAL4Act5C.PP fail to undergo cell-cycle arrest posterior to the second mitotic wave and continue the cell cycle. Cell death is significantly suppressed in the clones, even though abundant apoptosis is detected in neighbouring wild-type cells. Ommatidial clusters expressing ykiScer\UAS.cHa under the control of Scer\GAL4Act5C.PP have the normal complement of photoreceptor cells. The spacing between adjacent ommatidial clusters is increased due to the presence of extra interommatidial cells. The formation of extra interommatidial cells is most evident in pupal eye discs, where the clones contain many additional cells between photoreceptor clusters.

Clones expressing ykiScer\UAS.cHa under the control of Scer\GAL4Act5C.PP lead to marked overgrowth in adult epithelial structures. Wing imaginal discs containing multiple clones can reach up to eight times the area of control wing discs raised under identical conditions. Adult cuticles secreted by cells expressing ykiScer\UAS.cHa under the control of Scer\GAL4Act5C.PP display an unusual texture.

Wing imaginal disc cells expressing ykiScer\UAS.cHa under the control of Scer\GAL4Act5C.PP multiply faster than wild-type cells (a cell-doubling time of 12.0 hours compared to 16.1 hours in wild-type). These cells also minimize their contact with other cells and form round smooth cell borders. Cells expressing ykiScer\UAS.cHa under the control of Scer\GAL4Act5C.PP have a similar cell cycle profile and cell size distribution as compared to wild-type cells.

The co-expression of ykiUAS.cHa and pnutGD1512 from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] enhances the third instar larval wing disc dorsal compartment enlargement induced by the expression of ykiUAS.cHa from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts], even leading to the formation of tumors and to cell polarity defects (i.e. mislocalization of Discs large protein); this tumor phenotype is enhanced by the co-expression of bskDN.UAS. The pnutGD1512, ykiUAS.cHa co-expressing larvae fail to pupate and, instead, continue to grow to form giant larvae.

The co-expression of ykiUAS.cHa and pnutGD1512 under the control of either Scer\GAL4hh.PU or Scer\GAL4e22c leads to a much more severe enlargement of the wing disc posterior compartment as compared to the ykiUAS.cHa single expression condition; the Scer\GAL4hh.PU-driven co-expression also leads to some of the expressing cells moving and invading the wing disc anterior compartment.

The third instar larval wing disc dorsal compartment enlargement induced by the expression of ykiUAS.cHa from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts] is significantly enhanced by the co-expression of diaHMS00308, chbHMS01146, pblJF02979 or IncenpGD7511.

The co-expression of ykiUAS.cHa enhances the polyploidy/aneuploidy in the third instar larval wing disc dorsal compartment induced by the expression of pnutGD1512 from the second instar larval stage under the combined control of Scer\GAL4ap-md544 and Gal80[ts].

Co-expression of cactHMS00084 suppresses the reduced survival rates in response to Staphylococcus aureus (and the increase in bacterial load) seen in male flies with overexpression of ykiScer\UAS.cHa driven in the fat body by Scer\GAL4r4.

The increased number of satellite boutons on neuromuscular junctions in third instar larvae expressing StripdsRNA.shRNA.Scer\UAS.9 under the control of Scer\GAL4RapGAP1-OK6 is not suppressed by co-expression of ykiScer\UAS.cHa.

Moreover ykiScer\UAS.cHa clones induced earlier in development are never recovered at the end of larval development.

In dmPL35/+;dmαTub84B.PBb females, ykiScer\UAS.cHa overexpression clones are smaller than wild-type, whereas in males they are completely outcompeted by 48 hours after heat-shock.

Mosaic clones expressing hthGD4741 and ykiScer\UAS.cHa (under the control of Scer\GAL4αTub84B.PP) exhibit down-regulation of dm in the notum and do not grow tumors in that region. These clones are indistinguishable from ykiScer\UAS.cHa clones in the wing pouch region (where hth expression is not detected).

Co-expression of sdScer\UAS.cSa and ykiScer\UAS.cHa under the control of Scer\GAL4GMR.PF results in eyes which are severely overgrown (and much larger than the eyes of animals expressing ykiScer\UAS.cHa alone under the control of Scer\GAL4GMR.PF) and folded. There is a general loss of eye pigmentation in the posterior region of the eye.

The large eye phenotype caused by expression of ykiScer\UAS.cHa under the control of Scer\GAL4GMR.PF is suppressed by sd12/+.

The large size and round shape of clones in the eye disc expressing ykiScer\UAS.cHa under the control of Scer\GAL4tub.PU is suppressed if the clones are also homozygous for sd12.

Homozygous vg83b27-R clones that are also expressing ykiScer\UAS.cHa under the control of Scer\GAL4tub.PU are readily recovered in the wing blade region of the wing disc.

In clones with a Df(3L)banΔ1 background, the induction of extra interommatidial cells causes by expression of ykiScer\UAS.cHa, under the control of Scer\GAL4GMR.PF in a wild-type background, is partially suppressed. The Scer\GAL4GMR.PF>ykiScer\UAS.cHa, Df(3L)banΔ1 retinae have 3.1 cells in regions corresponding to one side of an ommatidial hexagon between two vertices while the number if 5.9 cells in Scer\GAL4GMR.PF>ykiScer\UAS.cHa retinae but 1.67 cells in wild-type retinae.

Df(3L)banΔ1 mutant clones in the wing that overexpress ykiScer\UAS.cHa under the control of Scer\GAL4tub do not grow to be as large as those that overexpress ykiScer\UAS.cHa in wild-type clones.
