Overexpression of Pink1Scer\UAS.T:Ivir\HA1 has no impact on ommatidia even at one month of age.

Overexpression of Pink1Scer\UAS.T:Ivir\HA1 using Scer\GAL4elav.PU has no impact on the longevity of flies.

Expression of Pink1Scer\UAS.T:Ivir\HA1 driven by Scer\GAL4GMR.PU results in a rough eye phenotype (disrupted ommatidial array organisation and bristle loss).

Flies expressing Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU do not exhibit any discernible eye phenotype, as compared to controls.

Flies expressing Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF exhibit a rough eye phenotype and disarrayed ommatidia.

Flies with overexpression of Pink1Scer\UAS.T:Ivir\HA1 driven by Scer\GAL4ey.PU have small eyes with disrupted ommatidial patterns.

Expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF or Scer\GAL4ey.PH results in a rough eye phenotype.

Expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Mef2.PR results in complete lethality.

Expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF causes a disorganisation of the ommatidial array and roughening of the external eye morphology.

Expression of Pink1Scer\UAS.T:Ivir\HA1 or parkScer\UAS.T:Zzzz\FLAG driven by Scer\GAL4GMR.PF does not suppress age-dependent retinal degeneration (loss of rhabdomeres from photoreceptors) in trpP365/+ flies; co-expression of both Pink1Scer\UAS.T:Ivir\HA1 and parkScer\UAS.T:Zzzz\FLAG results in significant partial suppression of the phenotype.

Co-expression of Pink1Scer\UAS.T:Ivir\HA1 and parkScer\UAS.T:Hsap\MYC using Scer\GAL4elav.PU is deleterious as no adult flies emerge from pupae.

Co-expression of TgA.Scer\UAS and Pink1Scer\UAS.T:Ivir\HA1 driven by Scer\GAL4GMR.PU results in enhanced rough eye phenotypes compared to expression of either construct alone. Co-expression of TgNIG.7356R partially suppresses the rough eye phenotype seen in flies with Pink1Scer\UAS.T:Ivir\HA1 driven by Scer\GAL4GMR.PU.

Expression of LonGD14030 enhances the disorganised eye phenotype seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4ey.PH.

Expression of LonHMS01060 enhances the disorganised eye phenotype seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4ey.PH.

Heterozygous LonG3998 suppresses the thoracic indentation phenotype seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4Mef2.PR.

Heterozygous Df(3L)Exel9011 suppresses the thoracic indentation phenotype seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4Mef2.PR.

Co-expression of parkScer\UAS.T:Hsap\MYC and Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF results in lethality.

Co-expression of Sirt1KK109370 or Sirt1GD11580 partially suppresses eye phenotypes seen in Scer\GAL4ey.PU>Pink1Scer\UAS.T:Ivir\HA1 flies.

Co-expression of Pink1Scer\UAS.T:Ivir\HA1 and parkScer\UAS.T:Hsap\MYC under the control of Scer\GAL4hs.PB at 25[o]C results in pupal lethality, but at 18[o]C, several adult escapers are obtained. These escapers have smaller mitochondria in the indirect flight muscles compared to controls.

The rough eye phenotype of flies expressing Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF or Scer\GAL4ey.PH is suppressed in a park25/park25 background.

The lethality phenotype of flies expressing Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Mef2.PR is partially rescued in a park25/park25 background.

The rough eye phenotype of flies expressing Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF is enhanced in the presence of Df(1)Exel6239/+ or Opa1s3475/+ mutations, or by co-expression of parkScer\UAS.cGa or MarfGD11094.

The rough eye phenotype of flies expressing Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF is suppressed in the presence of Df(2L)C144/+ or Drp1T26/+ mutations, or by co-expression of Drp1GD10456.

Expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4unspecified does not rescue the flight defects of park25/park25 mutants.

park25 partially suppresses the rough eye and ommatidial organisation phenotypes seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4GMR.PF. Addition of HtrA2Δ1 suppresses the phenotype completely.

Co-expression of Pink1Scer\UAS.T:Ivir\HA1 and parkScer\UAS.cGa under the control of Scer\GAL4GMR.PF results in a severe rough eye phenotype, which greatly exceeds that conferred when Pink1Scer\UAS.T:Ivir\HA1 is expressed alone. This phenotype is not suppressed in a HtrA2Δ1 mutant background.

Expression of HtrA2Scer\UAS.cPa enhances the disorganised eye phenotype seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4GMR.PF, resulting in a significant reduction in eye size. This phenotype is not suppressed in a park25 mutant background.

HtrA2Δ1 largely suppresses the rough eye phenotype seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4GMR.PF.

rho-7pΔ1 does not suppress the disorganised eye phenotype seen when Pink1Scer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4GMR.PF.

Expression of Pink1Scer\UAS.T:Ivir\HA1 enhances the rough eye phenotype seen when rho-7Scer\UAS.cWa is expressed under the control of Scer\GAL4GMR.PF. The eye is much smaller with distorted morphology and complete loss of bristles. A less severe rough eye phenotype is seen when Pink1Scer\UAS.T:Ivir\HA1 is co-expressed with rho-7SA.Scer\UAS.

The defective wing and thorax phenotypes seen in park1 mutant flies are not rescued by expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PB. Expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PB also fails to rescue the enlarged mitochondria and apoptotic muscle cells seen in adult park1 thoracic muscles.

Overexpression of Pink1Scer\UAS.T:Ivir\HA1 significantly reduces mitochondrial spheroid formation in photoreceptor neurons. Pseudo-pupil analysis reveal that Pink1Scer\UAS.T:Ivir\HA1 overexpression markedly reduces photoreceptor loss in 1-day-old flies.

Overexpression of Pink1Scer\UAS.T:Ivir\HA1 results in the reduction of the number of degenerative neurons showing cytoplasmic vacuolization in the olfactory lobes. Pink1Scer\UAS.T:Ivir\HA1 significantly increases both mean and median survival of flies expressing Hsap\HTTQ93.ex1p.Scer\UAS.

Overexpression of Pink1Scer\UAS.T:Ivir\HA1 in heterozygous park1 flies which also express Scer\GAL4elav.PU>Hsap\HTTQ93.ex1p.Scer\UAS fails to increase life span.

Overexpression of Pink1Scer\UAS.T:Ivir\HA1 in heterozygous park1 flies which also express Scer\GAL4elav.PU>Hsap\HTTQ93.ex1p.Scer\UAS fails to provide protection against photoreceptor degeneration.

Co-expression of MarfmiRNA.cUa.Scer\UAS results in a significant reduction of Pink1Scer\UAS.T:Ivir\HA1-mediated neuronal rescue in Scer\GAL4elav.PU>Hsap\HTTQ93.ex1p.Scer\UAS flies.

Overexpression of Pink1Scer\UAS.T:Ivir\HA1 in the presence of heterozygous porink05123 fails to extend lifespan or suppress neuronal loss in flies also expressing Scer\GAL4elav.PU>Hsap\HTTQ93.ex1p.Scer\UAS.

Co-expression of Hsap\UCP2Scer\UAS.cFa and Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU does not result in a rough eye phenotype.

Comments

The enlarged or aggregated mitochondria phenotype observed in the body wall muscles of Pink1B9 hemizygous male third instar larvae is restored by expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4how-24B.

Expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav.PU rescues the loss of dopaminergic neurons in the PPL1 cluster and adult thorax defects of Pink1B9 flies.

Expressing Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PB rescues the crushed thorax and downturned wing phenotypes seen in Pink1B9 mutant flies at both 3 and 45 days post-eclosion. The mitochondrial defects, reduction in ATP levels and flightlessness are also rescued. Expression of Pink1Scer\UAS.T:Ivir\HA1 suppresses the dopaminergic neuron loss in 45 day old Pink1B9 mutant flies.

Expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PB almost fully rescues the apparent behavioural phenotypes of Pink1B9 mutants. Similarly, impaired mitochondria in the indirect flight muscle of Pink1B9 mutants are fully recovered by Pink1Scer\UAS.T:Ivir\HA1 expression under the control of Scer\GAL4hs.PB.

The reduced climbing ability, flightless and downturned wing phenotypes seen in Pink1B9 flies are rescued by expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PB. The collapsed thorax phenotype seen in Pink1B9 flies is largely rescued by expression of Pink1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PB.
