Expression of NGD144 under the control of Scer\GAL4en-e16E leads to increased cell death in third instar larval wing discs.

The clonal expression of NGD144 under the control of Scer\GAL4Act.PU leads to several polar/stalk cell defects compared to controls: decreased number of polar/stalk cells; stalk cells do not show the normal intercalated morphology; and high frequency of fused egg chambers.

Expression of NGD144 under the control of Scer\GAL4ple.PU as well as the Scer\GAL4elav.PU driver in combination with Scer\GAL80ple.PK (all together with tub-Gal80[ts] to limit the time of expression to adulthood) results in significantly decreased adult lifespan.

Expression of NGD144 under the control of Scer\GAL4insc-Mz1407 results in the loss of type II neuroblasts.

Expression of NGD144 under the control of Scer\GAL4esg-NP7397 in the adult intestine (using the temperature sensitive Scer\GAL80ts.αTub84B allele to limit expression) at 22[o]C results in a mild increase in the density of both enteroendocrine cells and enterocytes, although the ratio of these two differentiated cell types is normal. At 25 and 28[o]C, there is a dramatic increase in the number of intestinal stem cell-like cells, as well as an increase in enteroendocrine cells and concomitant loss of enterocytes.

NGD144 driven by Scer\GAL4sev.PU leads to significant cone cell loss in third instar larvae eye discs, but no defects are observed in the photoreceptors.

Adult flies expressing NGD144 driven by Scer\GAL4sev.PU have glossy retinas and the eyes contain black patches of tissues indicating necrosis.

Adults expressing NGD144 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4Mef2.PR results in late pupal lethality.

Expression under the control of Scer\GAL4pnr-MD237 results in extra bristles on the notum in 10-20% of the Scer\GAL4pnr-MD237 expression domain.

Expression under the control of Scer\GAL4pnr-MD237 results in loss of bristles on the notum in 90-100% of the Scer\GAL4pnr-MD237 expression domain.

The co-expression of NGD144 (together with Dicer-2 transgene, for efficient RNAi) fully suppresses the increased number of type II neuroblasts in third instar larval brains resulting from the co-expression of lilliScer\UAS.SR.T:Zzzz\FLAG and Su(Tpl)Scer\UAS.SR.T:Hsap\MYC under the control of Scer\GAL4insc-Mz1407, even to the point of near absence of type II neuroblasts, as compared to controls; a similar suppression is not achieved when the phenotype arises from the expression of E(spl)mγ-HLHScer\UAS.cLb alone.

Co-expression of either MarfmiRNA.cUa.Scer\UAS or dmScer\UAS.cZa partially rescues the loss of type II neuroblasts seen in animals expressing NGD144 under the control of Scer\GAL4insc-Mz1407, while simultaneous co-expression of both MarfmiRNA.cUa.Scer\UAS and dmScer\UAS.cZa together fully rescues the phenotype.

wgl-17/+ suppresses the necrosis seen in adult eyes when NGD144 is driven by Scer\GAL4sev.PU.

wgl-17/+ suppresses the cone cell loss phenotype seen in the third instar eye discs when NGD144 is driven by Scer\GAL4sev.PU.
