Wild type third instar larval wing discs do not show cell death at 96h after egg deposition (AED), whereas Scer\GAL4bi.PU>Nop60BGD16822 discs show cell death along the A/P boundary (further increasing and spreading throughout the wing pouch at 120h AED); a large number of pyknotic nuclei are also detected basally in the pseudo-stratified epithelium. Scer\GAL4bi.PU>Nop60BGD16822 discs also show morphological shape changes (crumpled, folded, extensive reorganization and remodeling), with bending of the A/P boundary indicative of localized overgrowth.

Expression of Nop60BGD16822 driven by Scer\GAL4en.PU leads to over-proliferation in the posterior compartment of third instar larval wing discs.

Scer\GAL4bi.PU>Nop60BGD16822 adults have crumpled, highly disorganized blistered wings.

Epithelial-mesenchymal transition is induced in the peripodial membrane (cell fate changes) with silencing of Nop60BGD16822 driven by Scer\GAL4bi.PU in third instar larval wing discs.

Expression under the control of Scer\GAL41407 results in loss of neuroblasts in the larval brain associated with decreased size of the remaining neuroblasts.

Expression under the control of Scer\GAL41407 results in a reduction in the average size of neuroblasts together with shorter neuroblast lineages (less daughter cells per neuroblast) in the larval brain compared to controls.

Expression of Nop60BGD16822 in the whole wing blade and hinge under the control of Scer\GAL4nub-AC-62 results in a 45% average reduction in wing size. Only 10-0% of the small wings are correctly patterned, and most show moderate or severe developmental defects ranging from ectopic or irregular vein formation and wing blisters, to complete disorganisation of the wing blade which appears crumpled and vestigial. A decrease in wing size is accompanied by a decrease in cell size.

Expression of Nop60BGD16822 in the dorsal compartment of the wing disc, and more broadly throughout the developing wing pouch later (under the control of Scer\GAL4Bx-MS1096) results in severe wing malformation. Wings show absent or irregular margins, are often strongly underdeveloped and highly disorganised. Wing undergrowth is more marked in the dorsal compartment and the wing blades curve upwards. Lack of adhesion between the dorsal and ventral wing surfaces cause frequent formation of blisters.

Expression of Nop60BGD16822 in the wing at the dorsal-ventral boundary under the control of Scer\GAL4vg.int2.1 results in incomplete and notched wing margins with variable scalloping of the wing blade. Loss or irregular patterning of the margin bristles is also observed.

Expression of Nop60BGD16822 in the posterior compartment of the wing under the control of Scer\GAL4en.PU results in a significant reduction in this area of the wing, notches and loss of hairs and the posterior margin, and alternations in the positions of the posterior veins. 30% of flies show strong disorganisation of the whole wing blade. Cell size is normal in the posterior compartment. In the third instar larval wing disc, a significant increase in levels of apoptosis is observed compared to control tissue. No alteration in levels of cell proliferation is observed in the mutant tissue.

Expression of Nop60BGD16822 under the control of Scer\GAL4ap-md544 results in larval lethality. A small number of adult escapers display notum and/or wing defects. Reduced cell proliferation levels are observed in the dorsal section of the wing disc compared to the control ventral section. Large areas of apoptotic cell death is observed in cells derived from the dorsal wing disc.

Expression under the control of Scer\GAL4pnr-MD237 results in the absence of 80-90% of the Scer\GAL4pnr-MD237-expressing area of the notum.
