Expression of rkGD14383 RNAi under the control of Scer\GAL4slbo.2.6 leads to detachment defects during migration of the border follicle cell clusters: vast majority of cells within the cluster remain tethered to the anterior epithelium during initial migration attempts and some fail to leave the anterior altogether. Others do leave but maintain a tether (comprised of border cell membranes) to the epithelium which stretches and finally breaks as the cluster migrates. However, just over a half of the clusters manage to complete migration eventually. In contrast, expression of the RNAi in the inner polar cells of the cluster only using the Scer\GAL4upd1.PU has no significant effect on the cluster migration.

Expressing rkGD14383 in all border follicle cells using the Scer\GAL4slbo.2.6 driver together with Scer\GAL4upd1.PU results in migration defects and cluster tethering. The clusters also show mislocalization of E-cadherin and apical polarity markers.

Expressing rkGD14383 under the control of Scer\GAL4GMR.PU leads to a subtle rough eye phenotype.

Animals expressing rkGD14383 under the control of Scer\GAL4Act5C.PI (carried in the P{Act5C-GAL4}17bFO1 insertion line) and reared in standard growing conditions within food vials are unable to survive beyond the larval stage. The few third instar larvae that are recovered often die with a "double vertical plates" phenotype which is indicative of failed larval ecdysis.

Animals expressing rkGD14383 under the control of Scer\GAL4Act5C.PI (carried in the P{Act5C-GAL4}17bFO1 insertion line) which are allowed to mature on grape juice agar plates can develop until the pupal stage, at which point a range of lethal phenotypes is seen. Some pupae resemble larvae in shape. Pupae that arrest before or shortly after pupal ecdysis are seen and these pupae are pale in colour and extremely flimsy, suggesting that the puparia have not "tanned". Pharate adults are also seen, but they always fail to eclose.

Animals expressing rkGD14383 under the control of Scer\GAL4Act5C.PI (carried in the P{Act5C-GAL4}25FO1 insertion line) primarily die at the pupal stage. The few progeny which do eclose are unable to tan their cuticle or expand their wings. The flies also have great difficulty standing, and die within 24 hours of eclosion.

100% of animals expressing rkGD14383 under the control of Scer\GAL4T76 die prior to pupal ecdysis.

All animals expressing rkGD14383 under the control of Scer\GAL4T80 die as pharate adults.

Most animals expressing rkGD14383 under the control of Scer\GAL469B die as pharate adults. Adult escapers have untanned cuticle and unexpanded wings and the legs cannot support the body weight of the flies due to severe cuticular defects. The escapers die within 24 hours of eclosion. Animals which fail to eclose show abnormal leg shrugging.

Animals expressing rkGD14383 under the control of Scer\GAL4C855a eclose normally, but the adults show great difficulty walking and climbing, as manifested by dragging their metathoracic legs during locomotion. The metathoracic tarsi are kinked. The wings are expanded, but never become rigid, resulting in a sagging appearance.

Adults expressing rkGD14383 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in viable flies or early pupal lethality.

Expression under the control of Scer\GAL4pnr-MD237 results in a colour difference between the central Scer\GAL4pnr-MD237 expression domain of the notum and the surrounding lateral region in 0% or 100% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Expression under the control of Scer\GAL4pnr-MD237 results in the absence of 0% or 20-30% of the Scer\GAL4pnr-MD237-expressing area of the notum, depending on the insertion line used.
