Embryonic dopaminergic neurons in primary culture expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4ple.PF display resistance to rotenone-treatment induced lethality and mitochondrial fragmentation, and increased mitochondrial motility, as compared to control neurons.

Dopaminergic neurons in flies expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4ple.PF exhibit mitochondrial aggregation and fusion, as compared to controls.

Flies expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4GMR.PU do not exhibit any discernible eye phenotype, as compared to controls.

Flies expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4ple.PF and raised on rotenone show reduced loss of PAL, PPM1/2, PPM3 and PPL1 dopaminergic neurons compared to wild-type flies raised under the same conditions. The mutant flies also show better survival compared to controls when raised on rotenone.

The progressive loss of climbing ability seen when flies are raised on rotenone is ameliorated if the flies are expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4ple.PF.

Flies expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4ple.PF and raised under normal culture conditions have a modest but significant increase in median lifespan compared to controls.

Flies expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4ple.PF and raised under normal culture conditions have significantly improved climbing ability (from 15 to 30 days of age) compared to age-matched controls.

Flies expressing Hsap\UCP2Scer\UAS.cFa under the control of Scer\GAL4ple.PF and raised under normal culture conditions show no change in mitochondrial DNA/nuclear DNA ratio compared to controls.

Under fasting conditions, the hemolymph of Scer\GAL4Ilp2.PR Hsap\UCP2Scer\UAS.cFa flies has up to a 29% increase in circulating sugars (glucose + trehalose) compared to controls.

Scer\GAL4Ilp2.PR-mediated expression of Hsap\UCP2Scer\UAS.cFa extends adult lifespan and renders flies more resistant to oxidative and starvation stress compared to controls. Average egg laying activities and spontaneous physical activity are comparable to controls.

Scer\GAL4Ilp2.PR Hsap\UCP2Scer\UAS.cFa flies have an average of 26% increase in glycogen content but no noticeable increase in lipid content.

Expression of Hsap\UCP2Scer\UAS.cFa using Scer\GAL4elav.Switch.PO in the presence of RU-486 extends life span on average 28% in females and 11% in males. Life span is still extended when expression is induced on day 10 of adult life, whereas induction later than day 20 has little effect.

Expression of Hsap\UCP2Scer\UAS.cFa using Scer\GAL4Mhc.Switch.PO in the presence of RU-486 has no positive effect on life span.

Mitochondria from heads of Hsap\UCP2Scer\UAS.cFa, Scer\GAL4elav.Switch.PO long-lived flies exhibit increased state 4 respiration rates and a lower respiratory control ratio.

Mitochondria from heads and thoraces of Hsap\UCP2Scer\UAS.cFa, Scer\GAL4elav.Switch.PO long-lived flies exhibit a 40% decrease in hydrogen peroxide production and a 32% decrease in 4-hydroxy-2-nonenal (a measure of oxidative damage).

Hsap\UCP2Scer\UAS.cFa, Scer\GAL4elav.Switch.PO long-lived flies have an increased resistance to paraquat, but are more sensitive to other stresses such as starvation, compared to controls.

There is no difference in egg laying, spontaneous physical activity or climbing speed between long-lived Hsap\UCP2Scer\UAS.cFa, Scer\GAL4elav.Switch.PO flies and controls.

When Hsap\UCP2Scer\UAS.cFa is co-expressed in the Hsap\HTTQ93.ex1p.Scer\UAS, Scer\GAL4Eaat1.PR background, the reduced life span phenotype is suppressed.
