Expression of Lgr3GL01056 (together with UAS-Dicer2 to enhance RNAi efficiency) under the control of either Scer\GAL4αTub84B.PL or Scer\GAL4elav.PU (but not Scer\GAL4repo or Scer\GAL4Ptth.cMa) abrogates the growth inhibition of undamaged (anterior) tissues - eye discs - in third instar larvae (104 hr after egg laying, AEL) following targeted irradiation (limited to posterior tissues and administered in late second instar, at 80 hr AEL) which is observed in wild-type larvae and also strongly reduces the developmental delay the regenerative growth of damaged tissues normally produces. Lgr3GL01056 expression driven by Scer\GAL4GMR18A01 or Scer\GAL4phm.PO combined with Scer\GAL80elav.PY or Scer\GAL4nSyb.PS also prevents the growth inhibition of undamaged tissue in targeted irradiation larvae, however Scer\GAL4phm.PO-controlled expression does not decrease the irradiation damage-induced pupariation delay compared to controls.

In the absence of expression of tissue damage, expression of Lgr3GL01056 controlled by either Scer\GAL4αTub84B.PL or Scer\GAL4phm.PO does not inhibit eye disc tissue growth, with the Scer\GAL4αTub84B.PL driver as the discs are actually larger relative to age-matched controls.

Expression of Lgr3GL01056 under the control of either Scer\GAL4nSyb.PU, Scer\GAL4Lgr3.T:Hsim\VP16 or Scer\GAL4GMR19B09 results in a significant increase in fluctuating asymmetry of left and right wing areas compared to controls. Expression under the control of Scer\GAL4P0206 has no significant effect on fluctuating asymmetry.

Expression of Lgr3GL01056 under the control of Scer\GAL4tub.PU results in a mild reduction in wing disc size compared to controls.

Expression of Lgr3GL01056 under the control of Scer\GAL4αTub84B.PL, Scer\GAL4arm.PS, Scer\GAL4ChAT.7.4, Scer\GAL4phm.PU, Scer\GAL4GMR19B09, Scer\GAL4GMR17G11, Scer\GAL4Mz699 or Scer\GAL4elav.PU does not lead to any significant delay to pupariation, as compared to controls.

Expression of Lgr3GL01056 under the control of Scer\GAL4elav.PU leads to an increase in fluctuating asymmetry in adult wing area, as compared to controls.

Expression of Lgr3GL01056 driven by Scer\GAL4tub.PU (but not Scer\GAL4elav.PU or Scer\GAL4GMR9F04) accelerates pupariation.

Expression of Lgr3GL01056 driven by Scer\GAL4elav.PU leads to increased fluctuating asymmetry, with significant left-right variations in the size of wings.

The eye disc growth inhibition as well as the pupariation delay (in the absence of tissue damage) observed in third instar larvae expressing Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4αTub84B.PL are suppressed by co-expression of Lgr3GL01056 (together with UAS-Dicer2 to enhance RNAi efficiency).

Similar eye disc growth inhibition is also observed in larvae expressing NosScer\UAS.cUa driven by Scer\GAL4phm.PO and this phenotype also cannot be suppressed by co-expression of Lgr3GL01056.

Expression of Lgr3GL01056 in neurons under the control of either Scer\GAL4nSyb.PU, Scer\GAL4Lgr3.T:Hsim\VP16 or Scer\GAL4GMR19B09 completely suppresses the developmental delay caused by expression of Ilp8Ecol\lexAop.cCa in the imaginal tissues under the control of Ecol\lexArn-89-lexA.

Expression of Lgr3GL01056 under the control of either Scer\GAL4Lgr3.T:Hsim\VP16 or Scer\GAL4GMR19B09 completely rescues the reduction in wing disc size caused by expression of Ilp8Ecol\lexAop.cCa under the control of Ecol\lexArn-89-lexA.

Co-expression of Lgr3GL01056 suppresses the developmental delay seen in flies expressing Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4elav.PU.

Co-expression of Lgr3GL01056 suppresses, but co-expression of both Lgr3GL01056 and Scer\GAL80elav.PU fails to suppress, the developmental delay seen in flies expressing Ilp8Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4αTub84B.PL.

Co-expression of Lgr3GL01056 suppresses the delay in pupariation seen with Ilp8Scer\UAS.T:Zzzz\FLAG driven by Scer\GAL4tub.PU.
