UASt regulatory sequences drive expression of a mir-276a "sponge" sequence, which consists of ten repetitive mir-276a complementary sequences with mismatches at positions 9-12 separated by a variable four-nucleotide linker. The sponge sequence is inserted into 3'UTR sequence downstream of the EGFP coding region, to enhance stability.
Flies expressing Avic\GFPEGFP.Scer\UAS-mir-276aSP under the control of either Scer\GAL4elav.PU or Scer\GAL4c547 have a significantly defective naive odour avoidance response to 4-methylcyclohexanol (1 x 10[-3] v/v) compared to wild type.
Flies expressing Avic\GFPEGFP.Scer\UAS-mir-276aSP under the control of either Scer\GAL4Orco.PU, Scer\GAL4GH146, Scer\GAL4GH298, Scer\GAL4c747 or Scer\GAL4Aph-4-c232 have a normal naive odour avoidance response to 4-methylcyclohexanol (1 x 10[-3] v/v).
Flies expressing Avic\GFPEGFP.Scer\UAS-mir-276aSP under the control of Scer\GAL4elav.PU and raised at 29[o]C (to inactivate a Scer\GAL80ts.αTub84B allele which is present) show a significant impairment in naive odour avoidance response to 4-methylcyclohexanol when they are maintained at 29[o]C after eclosion. If the flies are shifted to 18[o]C after eclosion (resulting in repression of Scer\GAL4elav.PU by Scer\GAL80ts.αTub84B) a significant restoration of naive odour avoidance response to 4-methylcyclohexanol is seen.
Flies expressing Avic\GFPEGFP.Scer\UAS-mir-276aSP under the control of Scer\GAL4ey-OK107 have a normal naive odour avoidance response to 4-methylcyclohexanol (1 x 10[-3] v/v), 3-octanol (1.5 x 10[-3] v/v), and to benzaldehyde (0.5 x 10[-3] v/v). The flies also show a normal avoidance response to electric shock (60V).
Flies expressing Avic\GFPEGFP.Scer\UAS-mir-276aSP under the control of Scer\GAL4ey-OK107 show significantly reduced long term olfactory memory (using 3-octanol and benzaldehyde as the odours) measured 24 hours after spaced repetitive training, but memory measured 24 hours after repetitive massed training or short-term memory measured immediately after one training session is normal.
Flies expressing Avic\GFPEGFP.Scer\UAS-mir-276aSP under the control of Scer\GAL4c747 show significantly reduced long-term olfactory memory measured 24 hours after spaced repetitive training.
