The RMCE cassette of the Mi{MIC}dncMI03415 insertion has been replaced with a EGFP-FlAsH-StrepII-TEV-3xFlag multi-tag cassette. The insertion is in a coding intron of dnc and is predicted to protein trap the gene. The insertion is also within CR44886, not in a coding intron, and is not predicted to protein trap this gene.
dncMI03415-GFSTF.2 adults in which the level of dnc protein has been reduced by expressing slmbScer\UAS.T:Zzzz\vhhGFP4 (which targets GFP-tagged proteins for degradation) under the control of Scer\GAL4117Y using a 3-day temperature shift to 28[o]C starting at 1 day after eclosion show a significant reduction in performance index in an aversive olfactory learning assay compared to control flies. This learning defect is fully rescued after 48 hours at 18[o]C (which allows the levels of dnc protein to be re-established).
