Expression of Zzzz\AOXScer\UAS.cFa was expected to ameliorate the mutant phenotype of tko25t, as it has previously been shown to mitigate the toxicity of respiratory chain inhibitors and to rescue mutant and knockdown phenotypes associated with cytochrome oxidase deficiency, however it had no effect on the phenotype of tko25t mutants.

Flies expressing Zzzz\AOXScer\UAS.cFa under the control of Scer\GAL4βTub.Switch, in combination with RU486 feeding, are phenotypically indistinguishable from controls.

No dopaminergic neuron loss is observed when Zzzz\AOXScer\UAS.cFa is expressed under the control of Scer\GAL4ple.PF. Total ATP content is comparable to controls.

Flies expressing Zzzz\AOXScer\UAS.cFa under the control of Scer\GAL4da.G32 show increased resistance to mitochondrial respiratory chain inhibitors antimycin A and KCN compared with wild-type. Differences in survival between experimental and control flies are observed after only ten minutes of exposure to the drugs inhibiting either complex III or IV. After 24 h of exposure only flies expressing Scer\GAL4da.G32>Zzzz\AOXScer\UAS.cFa survive. However, no difference between Zzzz\AOXScer\UAS.cFa-expressing flies and controls is observed when a complex I inhibitor (rotenone) is employed. Using isolated mitochondria, it is observed that Scer\GAL4da.G32>Zzzz\AOXScer\UAS.cFa expression is able to support state 3 oxygen consumption in the presence of antimycin and KCN (in contrast to wild-type), but (similarly to wild-type) not in the presence of rotenone. Moreover, the expression of Zzzz\AOXScer\UAS.cFa decreased the level of mitochondrial reactive oxygen species (mtROS) in the presence of complex III or IV inhibitors, but not in the presence of an inhibitor of complex I.

Using isolated mitochondria, it is found that mtROS production in the absence of inhibitors is decreased (compared with controls) as a result of Scer\GAL4da.G32>Zzzz\AOXScer\UAS.cFa-expression.

Expression of Zzzz\AOXScer\UAS.cFa under the control of Scer\GAL4da.G32 does not significantly increase lifespan compared to wild-type.

Flies expressing Zzzz\AOXScer\UAS.cFa under the control of Scer\GAL4da.G32 show increased resistance to cyanide and to antimycin compared to wild-type controls.

Expression of Zzzz\AOXScer\UAS.cFa under the control of Scer\GAL4Act.PU suppresses the lethality and defects in mitochondrial respiration seen in Baldspot02281 mutants.

Co-expression of Cint\NDXScer\UAS.cGa and Cint\AOXScer\UAS.cFa driven by Scer\GAL4da.G32 results in partial lethality (significant decreases in the number of progeny) and significant delays in eclosion timing (slightly more than a day, and a further day with two copies of Cint\NDXScer\UAS.cGa) in male and female flies, compared to controls.

Expression of Zzzz\AOXScer\UAS.cFa under the control of Scer\GAL4βTub.Switch, in combination with RU486 feeding in either larval stages or adulthood, is unable to rescue the bang sensitivity or developmental delay of tko25t mutants.

Co-expression of Zzzz\AOXScer\UAS.cFa and Scer\NDI1Scer\UAS.cSa under the control of Scer\GAL4da.G32 in tko25t/tko25t mutants results in complete lethality, and in tko25t/+ mutants results in partial lethality.

Co-expression of Zzzz\AOXScer\UAS.cFa rescues pupal lethality in flies with expression of COX7AKK110870 driven by Scer\GAL4da.PU; surviving adults show progressive locomotor defects (climbing ability significantly reduced at 1 day old and more so at 14 days old) and bang sensitivity (significant increase in recovery time at 29[o]C at 1 day old and more so at 7 days old), compared to controls. Co-expression of Zzzz\AOXScer\UAS.cFa rescues pupal lethality in flies with expression of COX5BGD6659 driven by Scer\GAL4G14. Co-expression of Zzzz\AOXScer\UAS.cFa (but not Scer\NDI1Scer\UAS.cSa) partially rescues lethality in flies with expression of COX4KK107510 or COX5BGD6659 driven by Scer\GAL4elav-C155. Co-expression of Zzzz\AOXScer\UAS.cFa partially rescues shortened lifespan in surviving males with expression of COX5BGD6659 driven by Scer\GAL4elav-C155. Co-expression of Zzzz\AOXScer\UAS.cFa suppresses shortened lifespan in males with expression of COX4KK107510 driven by Scer\GAL4elav.PLu. Co-expression of Zzzz\AOXScer\UAS.cFa partially suppresses climbing defects and bang sensitivity in flies with expression of COX7AKK110870 driven by Scer\GAL4elav-C155. Co-expression of Zzzz\AOXScer\UAS.cFa partially suppresses the uninflated wing phenotype in flies with expression of levyKK108963 driven by Scer\GAL4elav-C155.

Expression of Scer\GAL4da.G32>Zzzz\AOXScer\UAS.cFa restores the progeny in sesB1 females to 15-20% of control crosses. The short lifespan of sesB1 flies is marginally improved by Zzzz\AOXScer\UAS.cFa expression. Zzzz\AOXScer\UAS.cFa-expressing males show a statistically significant increase in their lifespan or 3-4 days compared with that of controls, but the effect in females is negligible.

Expression of Zzzz\AOXScer\UAS.cFa completely suppresses the lethality seen when tamKK100485 is expressed under the control of Scer\GAL4Act5C.PU. The reduction in total ATP content is also suppressed.

Expression of Zzzz\AOXScer\UAS.cFa completely suppresses the dopaminergic neuron loss seen when tamKK100485 is expressed under the control of Scer\GAL4ple.PF.

Expression of Zzzz\AOXScer\UAS.cFa under the control of Scer\GAL4da.G32 does not suppress the male sterility, flight defects or defects in mitochondrial morphology seen in Pink1B9 flies.
