The endogenous Cirl locus (ORF, 5' intergenic region encoding the putative promoter, 5' and 3' UTRs) is replaced with an attP site; the w+* marker has been removed from CirlKO.attP.w+, leaving a single loxP site.

CirlKO.attP.w- maternal, zygotic mutant embryos showed delayed extension of the embryonic axis and reduced cell intercalation. Wing disc clone boundaries are smother than in control clones.
CirlKO.attP.w- mutant larvae have no structural or ultrastructural abnormalities in the larval pentascolopidial chordotonal organs (lch5) and do not have any differences in the action current frequencies induced in the lch5 neurons due to optogenetic activation (using Zzzz\ChR2.XXM.UAS.tdTomato expressed under the control of Scer\GAL4iav.PP and activated with 460-500 nm light), compared to controls; the responses of CirlKO.attP.w- mutant lch5 neurons in larvae to mechanical stimulation are significantly reduced, compared to controls, with no effect on their responses to temperature stimuli.
