The RMCE cassette in TI{CRIMIC.GT15}ari-1T-GT15 has been replaced with an intron phase 0 TI{2A-GAL4} DNA cassette that contains a splice acceptor site, a T2A peptide, sequence encoding the GAL4 driver and a polyA signal. The T2A sequence induces ribosome skipping, and thus this insertion acts as a gene trap (only only 73 residues of ari-1 are predicted to be translated), while allowing expression of the GAL4 driver under the control of endogenous ari-1 regulatory sequences.
