A GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain) has been inserted at the start codon within exon 2 of dsx. This results in the hemidriver being expressed under the control of endogenous dsx sequences, along with deletion of exon 2.

Inhibition (using Ctet\TeTxLCFRT.stop.UAS) of neurons targeted by the split-GAL4 pair of Scer\GAL4dsx-GAL4(DBD)::Zip- and Hsap\RELAAD.VT017258 expressing Scer\FLP1L.8xlexAop under the control of Ecol\lexAGMR24B02 in males results in much less sine song during courtship than controls, but a similar amount of pulse song, with similar inter-pulse interval, carrier frequency and amplitude to controls.
Acute optogenetic activation (using Cnoc\ChR1Cs.20xUAS.FRT.Venus with 660 nm (red) light activation) of neurons targeted by the split-GAL4 pair of Scer\GAL4dsx-GAL4(DBD)::Zip- and Hsap\RELAAD.VT017258 expressing Scer\FLP1L.8xlexAop under the control of Ecol\lexAGMR24B02 in males, in the absence of females, does not significantly induce courtship song.
