This trifunctional enzyme consists of two major domains: a N-terminal part, containing the methylene-THF dehydrogenase and the methenyl-THF cyclohydrolase activities and a larger formyl-THF synthetase domain.

It remains possible, given their map locations and mutant phenotypes, that "pug" and "dn" correspond to the same locus. pug is a different gene from Nmdmc; pug (at 86C) appears to encode the cytoplasmic form of methylenetetrahydrofolate dehydrogenase, Nmdmc (at 85C) appears to encode the mitochondrial form of methylenetetrahydrofolate dehydrogenase.
