CaMKII was pre-loaded with nucleotides in the presence of Ca[2+]/Cam before testing binding to CASK.

Interaction in vitro; bait produced as a recombinant fusion protein in bacterial system; produced as a recombinant fusion protein in baculovirus-infected Sf21 cell system.

CaMKII can bind to CASK in the absence of nucleotides, or when preloaded with ADP or AMP-PNP (a non-hydrolyzable ATP analog). However, pre-loading CaMKII with ATP inhibits binding to CASK. The interaction of CASK with kinase dead CaMKII is unaffected by nucleotide status.

Autophosphorylation of CaMKII disrupts its binding to CASK and to Cam.

CASK inhibits CaMKII activity in vitro.

Cam protects the CaMKII-CASK complex from dissociation upon the addition of ATP.
