From: Steven Marygold <steven.marygold@XXXX> To: flybase-help@XXXX Subject: RpS12 (CG11271) gene annotation problem Date: Thu, 02 Feb 2006 11:50:13 \+0000 Hi, I've noticed a problem with the annotation report of RpS12 (CG11271). There appears to be one aberrant EST (SD11723.5prime) that has resulted in the real RpS12 gene (that corresponds to the annotated RpS12-A, \-B and \-F isoforms, as confirmed by BLAST searches) being associated with a different gene (corresponding to the RpS12-D and \-E isoforms). (RpS12-C is a mixture of the two.) This other gene appears to be real as it has a handful of ESTs to support it (according to Gbrowse at least), though BLAST searches using the so-called RpS12-PD or RpS12-PE protein sequences fail to identify orthologous sequences. This annotation error has had the knock-on effect of several transposon insertions in the 'other gene' (the annotated C, D and E isoform sequences) being wrongly classed as RpS12 mutations. I guess there could be other knock-on effects also, such as the FB comments on which Dfs might disrupt the real RpS12 gene. Hope you can clear this up! Thanks for your time, Steven. \-- Steven Marygold Ph.D. Growth Regulation Laboratory (Rm 503), Cancer Research UK London Research Institute, 44 Lincoln's Inn Fields, LONDON, WC2A 3PX U.K. Tel: \+44 (0)20 7269 3351 Fax: \+44 (0)20 7269 3581
