From: Kevin Cook <kcook@XXXX> To: flybase-updates@XXXX Subject: UAS-Hsap\PFN1 constructs and insertions Date: Fri, 08 Feb 2008 09:42:08 -0500 ( 14:42 GMT) The following information accompanied stocks donated to the Bloomington Stock Center by Jieya Shao and Marc Diamond, University of California at San Francisco. P{UAS-Hsap\PFN1.D} is a P{UAST} construct of Human Profilin 1 (PFN1). P{UAS-Hsap\PFN1.D}2 is a homozygous viable and fertile, second chromosome insertion. P{UAS-Hsap\PFN1.S137A} is a similar construct encoding a mutant PFN1 protein with a S137A amino acid substitution. Likewise, P{UAS-Hsap\PFN1.S137D} encodes PFN1 with a S137D substitution. P{UAS-Hsap\PFN1.S137A}2 is a homozygous viable and fertile, second chromosome insertion. P{UAS-Hsap\PFN1.S137A}3 and P{UAS-Hsap\PFN1.S137D}3 are homozygous viable and fertile, third chromosome insertions. Neither the S137A nor S137D substitutions affect the binding of PFN1 to actin as assayed by immunoprecipitation. The S137D protein variant does not bind huntingtin, whereas the S137A variant does. __________________________________________________________ Kevin Cook, Ph.D. Bloomington Drosophila Stock Center Department of Biology http://flystocks.bio.indiana.edu Jordan Hall 142 Indiana University 812-856-1213 1001 E.
