From: lcherbas@XXXX Subject: FlyBase personal communication: miscellaneous transformed cell lines Date: October 16, 2008 11:37:28 AM GMT- 04:00 Origin of miscellaneous transformed cell lines Lucy Cherbas Drosophila Genomics Resource Center, Indiana University S2-MtN (FBtc0000154) is a stably transformed clonal line made from S2 (FBtc000006) by transformation with a mixture of p8HCO (FBrf0053391), a plasmid expressing methotrexate resistance, and pMtNMg (FBrf0053391), a plasmid expressing full-length Notch from a metallothionein promoter. The cell line was made as part of a collaboration between the Cherbas lab, the Muskavitch lab, and the Artavanis-Tsakonis laboratory. S2-Tub-wg (FBtc0000165) is a stably transformed population of Schneider's line 2 cells (FBtc0000999) containing a plasmid in which wg protein is expressed from a tubulin promoter; the line constitutively exports WG into the medium. The line was made and donated to the DGRC by Roel Nusse's lab; the information given here was obtained from Roel Nusse. I do not have details about the structure of the expression plasmid or the resistance marker. S2-Mt-GFP-alphaTub84B (FBtc0000168) is a stably transformed population of Schneider's line 2 cells (FBtc0000999) containing a plasmid in which a tubulin84B-GFP fusion protein is expressed from a copper-inducible metallothionein promoter. The cells carry a hygromycin-resistance marker. The cells were contributed to the DGRC by S. L. Rogers, along with the above description. I do not know details of the expression plasmid or the resistance marker.
