BAC clones were recombineered to introduce sequences encoding fluorescence or epitope tags at specific positions within genes to tag specific protein isoforms. The clones were integrated at specific attP sites by phiC31 integrase-mediated transformation. Though the constructs are named for the tagged gene, other genes are often present in the constructs. In the construct names, 'GFP.FPTB' represents combinations of epitope tags and peptide sequences: GFP.FPTB is a C-terminal superfolderGFP-FLAG-PreScission-TEV-BLRP tag combination.
