The following information accompanied stocks donated to the Bloomington Stock Center by Christian Lehner, University of Zürich. M{UAS-HygroR.L} is a UASt construct expressing the Escherichia coli hyg gene, which encodes hygromycin B phosphotransferase and confers resistance to hygromycin. The hyg sequence was derived from the pCoHygro plasmid. Addition of hygromycin to standard fly food (0.6 mg/ml final concentration) was observed to kill all control larvae but not those expressing M{UAS-HygroR.L} driven by Act5C-GAL4. M{UAS-BlastR.L} is a UASt construct expressing the Aspergillus terreus BSD gene, which encodes blasticidin S deaminase and confers resistance to blasticidin. The BSD sequence was derived from the pCoBlast plasmid. Addition of blasticidin to standard fly food (150 micrograms/ml final concentration) was observed to kill all control larvae but not those expressing M{UAS-BlastR.L} driven by Act5C-GAL4. M{UAS-PuroR.L} is a UASt construct expressing the Streptomyces alboniger pac gene, which encodes puromycin N-acetyltransferase and confers resistance to puromycin. The pac sequence was derived from the pCoPuro plasmid. Addition of puromycin to standard fly food (150 micrograms/ml final concentration) was observed to kill all control larvae but not those expressing M{UAS-PuroR.L} driven by Act5C-GAL4. These constructs were created to analyze the immortalization potential of different embryonic cell types. M{UAS-HygroR.L}ZH-86Fb, M{UAS-BlastR.L}ZH-86Fb and M{UAS-PuroR.L}ZH-86Fb are all miniwhite-marked insertions into the M{3xP3-RFP.attP}ZH-86Fb target site at 86F8, 3R:11808359 (R6).
