In third instar larvae, Lgr3ag5RA labels approximately 40 cell bodies in the embryonic/larval brain and approximately 140 cell bodies in the ventral nerve cord (VNC). Intense expression was observed in a single pair of neurons at the tip of the VNC, a single dorso-ventral pair of midline neurons in the thoracic segment and a pair of bilateral neurons in the larval pars intercerebralis. The pair of VNC neurons express the cholinergic marker ScerGAL4ChAT.7.4, the GABAergic marker ScerGAL4Gad1.3.098 and the glutamatergic marker ScerGAL4VGlut.PU while the dorso-ventral pair of thoracic midline neurons express ScerGAL4ChAT.7.4. The neurites of these latter neurons project centrally and then anteriorly towards the brain, where they branch at its base. The bilateral pair of neurons in the larval pars intercerebralis co-express ScerGAL4ChAT.7.4, ScerGAL4Gad1.3.098 and ScerGAL4VGlut.PU. These neurons have a single neurite branching ventrally towards the subesophageal ganglion and dorsally into highly arborized proximal termini which reach nearby insulin producing cells, whose neurites extend through this arborization. These bilateral neurons also associate with axonal projections from Ilp7-XP positive adult dorsomedial neurosecretory cells. A number of other neurons in the larval brain also express ScerGAL4ChAT.7.4. Approximately 10 neurons per hemisphere co-express ScerGAL4GMR19B09, including the pair of bilateral neurons in the larval pars intercerebralis. A further 60 or so neurons express ScerGAL4GMR19B09 in the VNC. The bilateral neurons in the pars intercerebralis also co-express ScerGAL4Mz699, as do neurons in the embryonic/larval subesophageal ganglion and ventral nerve cord.
