A κB element (5'-GGGGACTTTCC-3') from the mammalian immunoglobulin/HIV promoter is situated upstream of a canonical TATAA element. A Cavener element (CAAC) and an ATG translation initiation codon are situated downstream of the transcription start site, followed by an FRT cassette which contains mCherry (without its own ATG codon) and two tandem stop codons. A short sequence coding for a poly-glycine run is located downstream of the second FRT site. The pLUC coding sequence (minus its normal ATG codon) is present downstream, in frame with the poly-glycine run. When the FRT cassette is present, a fusion protein with amino acids encoded by one FRT site and the mCherry open reading frame is produced.
