A Ubiquitin promoter is fused upstream of an FRT stop cassette, followed by a bicistronic transcript that encodes two open reading frames separated by a 2A linker (this results in separate translation of each protein). The first ORF comprises a tkv cDNA (clone LD08886) into which the VHH(GBP6) coding sequence has been inserted (flanked by linker sequences that includes 2xTag:V5 in the upstream linker) in between residues A66 and R67. The second ORF comprises a put cDNA (clone LP03259) into which the VHH(GBP1) coding sequence has been inserted (flanked by linker sequences that includes 3xTag:FLAG in the upstream linker) in between residues G29 and S30.
