Amino acid replacement: G148R. Nucleotide substitution: Mutation of GGA to AGG.
Missense mutation: glycine replaced by arginine in a position that is completely invariant in known cdc2 homologs.

Lethal phase is during the larval/pupal stage. Homozygous embryos develop essentially normally. RP2 neuron duplication and large RP2 neurons are seen in only 0.8% of cases.
Die after pupariation at the larval-pupal interface when the allele is inherited from the male. Weak maternal antimorphic effect is seen: if the cdc2D57 allele is provided from the female, the lethality acts embryonically. Larvae take 1-2 days longer than wild type to reach the wandering stage. Imaginal discs are missing or rudimentary and larval brains are reduced in size. Endoreplication and polytenization of larval cells is normal, as are salivary gland size and intensity of DNA staining of their polytene nuclei.
