Description

Insertion in the 5' untranslated region.

P{PZ} insertion in intron 1 of the CT22741 transcript of Cka, 359bp downstream of the 5' end of the transcript and 53bp downstream of the end of exon 1. P{PZ} element is inserted in the same orientation as the gene.

Detailed Description

Embryos lacking both maternal and zygotic Cka function (derived from females carrying homozygous germ-line clones) die during embryogenesis with a dorsal open phenotype. They have head defects and lack the dorsal epidermis, as indicated by a large dorsal hole in the cuticle. Embryos lacking both maternal and zygotic Cka function do not undergo the cell shape changes that normal occur in the leading edge and lateral epithelial cells during dorsal closure.

Cka05836/Ckas1883 mutant eye discs display a variable reduction in disc size. In some discs overall morphology appears normal. Malformed and misfolded discs are also observed, with a higher frequency in smaller discs.

Homozygotes die during the larval and pupal stages. Embryos derived from homozygous female germ line clones have a dorsal open phenotype which can be fully paternally rescued.

Germline clones produce eggs with patterning defects: head and dorsal open. Lethality occurs during the larval and pupal stages.

The weak dorsal-open phenotype seen in homozygous kay2 embryos is enhanced by Cka05836/+. The weak dorsal-open phenotype seen in homozygous bsk1 embryos is enhanced by Cka05836/Cka05836. The dorsal open phenotype seen in homozygous Cka05836 embryos derived from females carrying homozygous Cka05836 germ-line clones is substantially rescued by expression of JraAsp.hs.sev using heat shock. The dorsal open phenotype seen in homozygous Cka05836 embryos derived from females carrying homozygous Cka05836 germ-line clones is substantially rescued by expression of hepCA.Scer\UAS under the control of Scer\GAL4hs.PB using heat shock. The dorsal open phenotype seen in homozygous Cka05836 embryos derived from females carrying homozygous Cka05836 germ-line clones is substantially rescued by expression of tkvQ199D.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.PB using heat shock.

The P{EP}CkaEP2514 insertion is 1bp away from the P{PZ}Cka05836 insertion.

Precise excision of the insertion reverts the lethal mutant phenotype.
