Recordings from larval neuromuscular junctions (muscle 6 of A3 or 4) of rtP/Df(3L)ED4470 animals show a dramatic decrease in evoked synaptic transmission. However, the size and frequency of spontaneous mini excitatory junction potentials is normal. Based on this, estimated quantal content is reduced about 3 fold. This phenotype is unaffected by the suppression of action potentials with 1 micro-molar TTX. Short term facilitation is enhanced in these mutants, suggesting that decreased quantal content is due to decreased probability of vesicle release in response to stimulus.
The bodies of first instar larvae are rotated around the long axis, forcing the animals to move in a clockwise circle as they crawl. The larval cuticle presents a perfect segmental alignment of landmarks but the adult abdominal cuticle segments appear staggered relative to each other.
Homozygous males and females have reduced fertility and show abdominal rotation.

Evoked excitatory junction potentials are normal at the neuromuscular junctions of rtP/+ or Dge01554/+ larvae, but siginficantly reduced compared to wild-type in rtP/+; Dge01554/+ larvae. However, the size and frequency of spontaneous mini excitatory junction potentials is normal in transheterozygotes.
The rotated abdomen phenotype of rtP/rtEP571 flies is not enhanced by the presence of the Dp(1;Y)y2sc duplication. Likewise, expression of the twScer\UAS.T:Ivir\HA1 transgene, under the control of Scer\GAL4Act5C.PI, does not enhance the abdominal phenotype of rtP/rt2 mutants. This phenotype is also not affected by tw RNAi, achieved by expression of twdsRNA.Scer\UAS under the control of Scer\GAL4Act5C.PI. However, expression of twdsRNA.Scer\UAS, driven by Scer\GAL4Act5C.PI or Scer\GAL4αTub84B.PL, reduces the rotated abdomen phenotype of rtP/+ flies.
At 18oC, flies expressing Pomt2dsRNA.Scer\UAS under the control of Scer\GAL4Act5C.PHb in a rtP/+ background show a clear twisted abdomen phenotype, in contrast to either single mutant (rtP/+ or flies expressing Pomt2dsRNA.Scer\UAS under the control of Scer\GAL4Act5C.PHb at 18oC in a wild-type background).
