dppe90 homozygotes, kept at 18'C until 42 hours APF (equivalent to 20 hours APF at 25'C) and then raised to 25'C for the proceeding 22 hours have slightly abnormal arrangments of inter-ommatidial cells including: 3's which fail to establish a correct position as a vertex of the hexagon; 2'/3's abnormally arranged around sensory bristles; aberrant bristle-bristle contacts.

Females carrying temperature sensitive combinations dppe90/dpphr56 or dpphr4/dpphr56 after shifting to the restrictive temperature for one week show 40-50% of germaria that are significantly smaller than wild type. The effect is more pronounced in older females. Stem cell number is reduced, stem cells have differentiated into cysts. Some effect on egg chamber budding is seen.
Embryos show cuticular holes, disorganization of the head skeleton, internalization of the seventh and eighth abdominal segments and the filzkorper. dppe90 shows a temperature sensitive embryonic lethality: percentage embryonic lethality drops from 29% at 25oC to 15% at 18oC. Temperature sensitivity is evident in denticle pattern as well as survival.
Homozygous and transheterozygous embryos were examined with respect to 25 cuticular markers, results demonstrate a graded requirement for dpp along the dorso-ventral axis.
dppe90/dpp[+] is a suppressor | partially of increased cell number | female | adult stage phenotype of PscGD4481, Scer\GAL4C587, Su(z)2GD16388
dppe90/dpp[+] is a suppressor | partially of neoplasia | female | adult stage phenotype of PscGD4481, Scer\GAL4C587, Su(z)2GD16388
dppe90/dpp[+] is a suppressor | partially of germarium phenotype of PscGD4481, Scer\GAL4C587, Su(z)2GD16388
dpphr56/dppe90, gbbD4 has male germline stem cell phenotype
dpphr56/dppe90, gbb[+]/gbbD4 has male germline stem cell phenotype
Simultaneous expression of both PscGD4481 and Su(z)2GD16388 under the control of Scer\GAL4C587 (together with tub-Gal80[ts] to limit the time of expression to adulthood) results in tumorigenic accumulation of spectrosome containing germline stem cell-like (GSC-like) cells (which increase in number with the time of expression) in female germaria; this accumulation of GSC-like cells is significantly suppressed by combination with a single copy of dppe90.
bamhs.PO animals cultured at 18oC and heat shocked as late stage pupae (37oC, 4 times for 2 hours each with an 8-12 hour interval) produce adult females with agametic germaria. If these animals also carry the temperature sensitive allelic combination dpphr56/dppe90, and are shifted to 29oC (the non-permissive temperature) for 10 days, the resulting agametic germaria contain only slightly less follicle cells than controls (bamhs.PO only).
Nusslein-Volhard.
Based on considerations of degree of dominant lethality, dpp alleles can be placed in an allelic series. Progressing from weakest to most severe the series is: dppe87 < dppe90 < dpphr56 < dpphr4 < dpphr92 < dpphr27 < dpphr93 < dppH94 < dppH95 < dppH96 = dppH91 = dppH46.
dppe90 shows interallelic complementation at 18oC with dpphr56 and dpphr4.
