Construct: slp1 gene expression is driven by a heat shock promoter.
Ectopic expression of slp1hs.PC using heat shock between 2.5 and 4.0 hours of development (at 22oC) results in additional GMC-1 -> RP2/sib lineages. Duplication of the RP2/sib lineage is seen in 18% of hemisegments and in 17% of hemisegments triplication of the lineage is seen. Additional neuroblasts, such as NB5-3, NB3-5 and NB5-6 have adopted an NB4-2 identity in these embryos. NB5-3 is the most frequently transformed neuroblast (approximately 35% of hemisegments), with the highest frequency of transformation seen when expression of slp1hs.PC is induced between 3.0 and 3.5 hours of development.

Expression of slp1hs.PC in wgl-8 or wgl-17 embryos rescues the RP2/sib lineage in as many as 85% of hemisegments. Rescue of NB4-2 is seen in approximately 43% of hemisegments. The RP2/sib lineage is missing in 97% of hemisegments in embryos carrying two copies of gsbhs.PBH and one copy of slp1hs.PC, as is seen in embryos carrying only gsbhs.PBH. In contrast, in embryos containing either one copy of gsbhs.PBH and one copy of slp1hs.PC or two copies of gsbhs.PBH and two copies of slp1hs.PC, both loss and gain of the RP2/sib lineage is seen in a partially penetrant manner.
