The increase in survival rate with Mmus\Pkacahs.PJ is larger at 22[o]C than at 18[o]C (~10% at 22[o]C and ~5% at 18[o]C).
Mmus\Pkacahs.PJ completely rescues the learning defect of Nf1P1 when the flies are raised at room temperature. Mmus\Pkacahs.PJ partially rescues the learning defect of Nf1P2 when the flies are raised at room temperature, and completely rescues Nf1P2 when it is expressed using heat shock (37oC, 30 minutes). Nf1P1 and Nf1P2 flies show a short-term memory defect (3 and 8 hour retention) which is completely rescued by heat shock induced expression of Mmus\Pkacahs.PJ.
Weak heat induced expression at 28oC causes significant rescue of the Nf1 mutant pupal phenotype. Higher expression levels cause lethality.
Mutated form of the catayltic subunit does not bind the regulatory subunit with high affinity and hence is constitutively active, irrespective of cAMP concentration.
