Salivary glands from first, second and early third instar larvae are resistant to expression of rprhs.PW, surviving normally and showing no staining for active caspase-3. In contrast, salivary glands from animals after the mid-third instar larval transition are susceptible to cell death induced by rprhs.PW.

Expression of rprhs.PW in S2 cells leads to increased cell death.

Third larval instar wing discs expressing rprhs.PW (using heat shock) show a high level of apoptosis.

Expression of rprhs.PW in S2 cells induces cell death.

Causes apoptosis leading to organismal death in the embryo, when heat shock is applied at 5-10 hours after egg laying.

Upon heat shock causes massive cell death throughout embryos. This cell death can be also be induced in a Df(3L)H99 background. A single heat pulse at a variety of stages is sufficient to kill flies, though late embryos and late pupae (where the rate of cell division is low) are more resistant to this lethality than other stages. 4-6 hours after a 1 hour heat shock at 39oC, half the flies carrying the heat inducible rpr died, suddenly.

Can induce apoptosis in Df(3L)H99 embryos.

Sufficient to induce extensive apoptosis increase in Df(3L)H99 embryos.

The high level of apoptosis caused by expression of rprhs.PW in the third larval instar wing disc is not affected if the discs are also mutant for Ada2bunspecified.

The cell death induced by rprhs.PW in S2 cells is blocked almost completely by treating the cells with Nc dsRNA (produced by annealing NccIa and Nca.cIa RNA). The cell death induced by rprhs.PW in S2 cells is partly suppressed by treating the cells with Ice dsRNA (produced by annealing IcecIa and Icea.cIa RNA), Dcp-1 dsRNA (produced by annealing Dcp-1cIa and Dcp-1a.cIa RNA) or Ark dsRNA (produced by annealing ArkcIa and Arka.cIa RNA).

Organismal death in the embryo, caused by rprhs.PW when heat shock is applied at 5-10 hours after egg laying is suppressed by thSL. thSL increases the viability of rprhs.PW flies threefold.
