Detailed Description

Cyt-b5k14202/Cyt-b5k09716 females and males produce few progeny, even when mated with wild-type flies, and the mutant females lay few eggs.

Cyt-b5k14202/Cyt-b5k09716 larvae contain large numbers of circulating lamellocytes (these are not found in normal larvae). Lamellocytes are more prevalent in larvae containing small melanotic tumours, while plasmatocytes are more abundant in larvae containing large melanotic tumours.

The number of plasmatocytes from both hematopoietic waves are increased in heterozygous larvae of both sexes compared to wild type.

Homozygous larvae exhibit melanotic tumours and aberrant fat body and gut.

The melanotic tumour phenotype seen in Cyt-b5k14202/Cyt-b5k09716 animals is rescued by expression of Cyt-b5Scer\UAS.cKa under the control of Scer\GAL4He.PZ, but is not rescued when expression is driven by Scer\GAL4Sgs3.PD.

The lethality of Cyt-b5k14202/Df(2R)ST1 animals is not rescued by expression of Cyt-b5Scer\UAS.cKa under the control of Scer\GAL4He.PZ, but is rescued when expression is driven by Scer\GAL4da.G32.

Comments

Mobilisation of P{lacW} reverts the lethal phenotype demonstrating the insertion causes the mutant phenotype. Level of Ecol\lacZ expression and viability of adults is not altered by bacterial infection of adults.
