Animals expressing bab24.Scer\UAS under the control of Scer\GAL4p339 or Scer\GAL430A have decreased pigmentation in the abdominal cuticle and the claw organs. Animals expressing bab24.Scer\UAS under the control of Scer\GAL471B or Scer\GAL4bab1-Pgal4-1 show incomplete fusion of the tergites. Animals expressing bab24.Scer\UAS under the control of Scer\GAL430A, Scer\GAL471B or Scer\GAL4p339 have wing defects. Animals expressing bab24.Scer\UAS under the control of Scer\GAL4E132 have malformed and branching legs, show a transformation of the basal cylinder and arista to tarsus and have a duplication of the haltere. The wings are shortened and broadened compared to wild type and there is an outgrowth in the hinge region of the wing. Wing discs of these animals show deformations and massive cell degeneration.
Males and females expressing bab24.Scer\UAS under the control of Scer\GAL4hs.PB at 25oC lack dark pigmentation in segment A6. Little of the dark pigmentation is left in A5 and A4. Flies carrying bab24.Scer\UAS under the control of Scer\GAL4hs.PB which are raised at 18oC then shifted to 32oC during the late third larval instar/pupal stage show a "split-tergite" phenotype (as well as loss of pigmentation); the tergite primordia of all abdominal segments do not fuse (this phenotype is normally only seen in A7). Animals expressing bab24.Scer\UAS under the control of Scer\GAL4hs.PB at 25oC show reduced pigmentation in A5 and A6.
Expression driven by Scer\GAL4pnr-MD237 in males causes loss of both male specific and non-sex-specific striped abdominal pigmentation. Expression driven by Scer\GAL4C-765 in males causes transformation of A6 and A7 to a more anterior identity. A7 tergite and sternites are restored. A6 sternite and tergite shape, bristles and trichomes resemble those of A5. There is no effect on the genitalia or analia of either species.
Expression of bab24.Scer\UAS under the control of Scer\GAL4bab1-Pgal4-2 fully rescues the mutant leg phenotypes of bab2E4/Df(3L)babAr07 flies; tarsal segments 2 and 3 do not have any sex combs and tarsal segments 4 and 5 are always properly separated. bab2E4/Df(3L)babAr07 females have ovaries of reduced size which contain only a few follicles with abnormal orientation. They are subfertile and lay very few eggs. These defects are fully rescued by bab24.Scer\UAS expressed under the control of Scer\GAL4bab1-Pgal4-2. bab2E4/Df(3L)babAr07 females show increased abdominal pigmentation; segments A5 and A6 are fully pigmented. This defect is not rescued by bab24.Scer\UAS expressed under the control of Scer\GAL4bab1-Pgal4-2.
The effect of Scer\GAL4pnr-MD237 in males causing loss of both male specific and non-sex-specific striped abdominal pigmentation is enhanced by abd-A,Abd-B deficiencies and partly suppressed by abd-A,Abd-B duplications. The effect of Scer\GAL4C-765 in males causing transformation of A6 and A7 to a more anterior identity is enhanced by abd-A,Abd-B deficiencies and partly suppressed by abd-A,Abd-B duplications.
