UASt regulatory sequences drive expression of Atpα with the amino acid replacement D369N. The nucleotide substitution that generates the amino acid change also results in the formation of a KpnI site.

Overexpression of AtpαD369N.Scer\UAS with Scer\GAL4C380 results in an increase in the resting membrane potential and the fire-rate response of larval motorneurons. Expression of AtpαD369N.Scer\UAS with Scer\GAL4P2.4.Pdf results in a reduction in total sleep and an increase in sleep latency during the day and night, compared to controls.

Animals expressing AtpαD369N.Scer\UAS under the control of Scer\GAL4nrv1.PS die by the late first larval instar. Animals expressing AtpαD369N.Scer\UAS under the control of Scer\GAL4nrv2.PS are more sensitive to ouabain than wild-type animals at all concentrations of ouabain tested. Adults expressing AtpαD369N.Scer\UAS under the control of Scer\GAL4nrv2.PS show temporary paralysis or uncontrolled wing beating after brief mechanical stimulation.
