The FRT>HcRed1>FRT cassette has been excised, allowing the UASt regulatory sequences to drive expression of an inverted repeat of a 500bp apolpp fragment (corresponds to the N-terminal 500bp of the coding sequence, which encodes part of the ApoLII protein produced by cleavage of the pro-Apolipophorin precursor).
Expression of RfabgdsRNA.ApoLII.Scer\UAS in the fat body under the control of Scer\GAL4Adh.PF (initiated by heat shock during the larval stages) results in a dramatic enlargement of the neutral fat droplets in the third instar larval posterior midgut.
When RfaBpApoLII.dsRNA.Scer\UAS is driven by Scer\GAL4αTub84B.PL (after heat shock driven FLP out of the marker cassette in RfaBpScer\FRT.ApoLII.dsRNA.Scer\UAS) animals prolong their third instar and rarely pupariate. When RfaBpApoLII.dsRNA.Scer\UAS is driven by Scer\GAL4Adh.PF (after heat shock driven FLP out of the marker cassette in RfaBpScer\FRT.ApoLII.dsRNA.Scer\UAS) causes a dramatic expansion of the lipid droplets in the posterior midgut. Lipid droplets in mutant discs are fewer and smaller than in wild-type. Their wing discs are also smaller in size, particularly in the wing pouch.
