UASt regulatory sequences drive expression of an inverted repeat.
Scer\GAL4Dot.PK-mediated expression of CubnGD6458 reduces the normal accumulation of ANF-RFP (expressed via Rnor\ANFMhc.T:Disc\RFP) in pericardial nephrocytes.
Uptake of a secreted marker protein by pericardial nephrocytes is blocked in animals expressing CubnGD6458 under the control of Scer\GAL4Dot.PK. The pericardial nephrocytes show ultrastructural defects compared to wild type: the number of endocytic vesicles and vacuoles are dramatically reduced, electron-dense lysosomes are nearly absent, nephrocyte diaphragms are reduced in number and lacunar structures are shortened.
Animals expressing CubnGD6458 under the control of Scer\GAL4Dot.PK are viable under normal conditions, but show dramatically reduced viability compared to controls in the presence of silver nitrate.
Expression via P{GAL4-tim.E}27 or P{cry-GAL4.Z}17 (in the presence of Dcr-2Scer\UAS.cDa) does not significantly affect circadian rhythmicity in locomotor activity assays in constant darkness.
Expression under the control of Scer\GAL4pnr-MD237 results in a colour difference between the central Scer\GAL4pnr-MD237 expression domain of the notum and the surrounding lateral region in 0% or 20-30% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.
The block in uptake of a secreted marker protein by pericardial nephrocytes caused by expression of CubnGD6458 under the control of Scer\GAL4Dot.PK is not rescued by co-expression of AmnionlessScer\UAS.cZa.
