Expression of Nab2GD11745 under the control of Nab2GD11745 leads to climbing defects in adult flies.

Expression of Nab2GD11745 (along with UAS-dicer2) driven by Scer\GAL4elav-C155 leads to ellipsoid body defects (lack a completely closed ring structure, with an opening left on the ventral side) and mushroom body defects (similar to those seen in Nab2ex3/Nab2ex3 flies: thinned/absent alpha/beta-lobes, variable degrees of beta-lobe fusion) in the adult brain, compared to controls. Mushroom body defects are more frequent in males than females. Expression of Nab2GD11745 driven by Scer\GAL4elav-C155 does not affect suppression of courtship index (in a courtship conditioning assay), showing learning is similar to controls; however, short-term memory is defective in Scer\GAL4elav-C155>Nab2GD11745 flies (trained flies show similar courtship index to sham flies). Locomotor activity in Scer\GAL4elav-C155>Nab2GD11745 flies is normal.

Expression of Nab2GD11745 under the control of Scer\GAL4elav.PU results in flight defects and in severe locomotion defects in a negative geotaxis assay.

Expression of Nab2GD11745 under the control of Scer\GAL4OK6 results in modest locomotion defects in a negative geotaxis assay.

Expression of Nab2GD11745 under the control of either Scer\GAL4Mef2.PR or Scer\GAL4Mhc.PW does not result in locomotion defects in a negative geotaxis assay.

Adults expressing Nab2GD11745 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

The locomotion deficit (decreased climbing ability) characteristic for adult flies expressing Nab2GD11745 under the control of Scer\GAL4elav-C155 is worsened further by combination with a single copy of Fmr1Δ113M but also by co-expression of Fmr1Scer\UAS.cWa (which on its own also induces climbing ability defects).
